Abstract

Objective: The purpose of this project is to study catechins on human hepatoma cell radiosensitivity, and further explore the mechanism. Focus on solving the following problems: (1) whether the catechins can increase the human hepatoma cell radiosensitivity of different concentrations of catechin sensitizing effects. (2) To investigate the mechanism of catechin radiosensitization. Methods: (1) cultured in vitro in human hepatocellular carcinoma cell line HepG2 as target cells using trypan blue staining and MTT assay explore catechin on HepG2 cells after irradiation by 2Gy survival and colony formation method study of catechin on the radiosensitivity of two human hepatocellular carcinoma cell lines HepG2 and BEL-7402 the fitting cell dose survival curve, calculated cells to ionizing radiation the sensitivity parameter D , D < sub> q , SF 2 , α and beta, calculate the 10% survival level catechin sensitizing Ser. (2) two human hepatocellular carcinoma cell line HepG2 and BEL-7402 treatment with catechin accept 5GyX ray irradiation at different times of the proportion of apoptosis was measured by HE staining and flow cytometry, with irradiation alone 5Gy the cell apoptosis levels were compared; HepG2 and BEL-7402 cells bcl-2 and Bax protein expression level changes by immunohistochemistry and flow cytometry determination of catechins. Results: (1) trypan blue staining and MTT assay showed that: catechins can reduce cell viability in HepG2 cells by 2Gy irradiation, and this role and the role of the concentration of catechins, time correlated. Colony formation ability of EGCG can increase the radiosensitivity of HepG2 and BEL-7402 cells, 1,10 microg / ml catechin two cells D 0 d q , SF 2 , α and beta decreased 1,10 microg / ml catechin HapG2 at SER were 1.25 and 1.54; 10% survival level; 1,10 microg / ml catechin SER BEL-7402 were 1.21 and 1 Stone 2. Door) HE staining and flow cytometry determination HepGZ Wo BEL-7402 level of apoptosis the SGy after irradiation, the results show two accept SGy irradiated cells treated with 1,10 ug / ml catechin, high rate of apoptosis in a separate accept the SGy exposure the apoptosis rate; immunohistochemistry and flow cytometry test showed that catechins after handling HepGZ BEL Diao 402 cells were lower than on the protein expression of Bax protein increased expression. Conclusion: * V catechin two people can increase the hepatoma cell HepGZ and BEL Diao 402 radiosensitivity. c plant tea prime increase HepGZ and BELJ402 cell radiosensitivity may be apoptosis rate increased cell irradiation, the molecular mechanism of catechins by changes in intracellular bclZ, Bax protein expression, so that the susceptibility to apoptosis increase, an increase in apoptosis in cells after irradiation.
