Abstract


Sensory neuron -specific receptor (sensory neuron-specific receptor, SNSR) specifically located in the dorsal root ganglia ( dorsal root ganglia , DRG ) and trigeminal ganglion (trigeminal ganglia, TG) neuron , class of neurons involved in the transmission of pain information . Biochemical experiments of this study, Western blot method SNSR role of morphine tolerance , is designed to explore the the SNSR modulation of morphine tolerance mechanism cytology level . The results showed that: the continuous injection of morphine 6 days resulted in upregulation of protein kinase C in the spinal cord and DRG ( proteinkinase C , PKC ) and neuronal nitric oxide synthase (neuronal nitric oxide synthesis, nNOS) , the TRPV1 expression had no effect ; every day morphine and the next day joint injection SNSR agonist bovine adrenal medulla hormone 8-22 ( bovine adrenal medulla 8-22 BAM8 - 22 ) can inhibit the generation of morphine tolerance , PKC and nNOS expression was significantly decreased TRPV1 expression unchanged. This study shows that intermittent activation the SNSR to inhibit morphine tolerance , its mechanism may be associated with PKC , NO signaling pathway and the TRPV1 pathway is relevant to be further research .
