Abstract

Purpose and significance of Yunnan Province, is located in the Southwest, the case in most parts of the tropical and subtropical climates, the complexity of the natural conditions, suitable for the distribution and dissemination of the insect vector and arbovirus previous survey confirmed Yunnan Province, there are nearly 10 species of insect-borne virus The distribution and prevalence of its related diseases. In addition to the wide range of distribution of Japanese encephalitis (Japanese encephalitis, JE), there are other arbovirus disease prevalence. Yunnan border areas often viral encephalitis and the incidence and prevalence of patients with unexplained fever. As many viral encephalitis and unexplained febrile illness arbovirus infection related, master arbovirus epidemic situation of the region, the subject is collected in some areas of Yunnan Province in serum and cerebrospinal fluid of viral encephalitis and unexplained febrile illness specimens related arbovirus nucleic acid detection and virus isolation, the related arboviruses positive samples, the determination of the gene sequence analysis and viral genotype study and application of molecular biology techniques. Through research, in order to grasp the arbovirus disease epidemic and pandemic strains, molecular epidemiological characteristics, and to provide a scientific basis for the region for Disease Control and Prevention. 2 July 2008 to September in the western and southern Yunnan Baoshan, Dehong, Dali and Xishuangbanna border areas to conduct investigations, collecting serum and cerebrospinal fluid specimens for viral encephalitis and fever of unknown origin, and individual cases survey, the establishment of the specimen collection and assay database, collected patient samples by RT-PCR and real-time PCR method arbovirus nucleic acid detection, virus isolation by C6/36 cell culture and suckling mice inoculated methods of these specimens . Clastal × 1.83 nucleotide sequence alignment the DNAstar the middle MegAlin Software nucleotide sequence and amino acid sequence homology analysis. The and turn JE the PrM-C section Dengue virus PrM-C, the NS5 section nucleotide sequence phylogenetic tree analysis using MEG4 software. 3 results in the above-mentioned 4 City has collected a total of 106 copies of the viral encephalitis and unexplained fever patient serum and CSF samples, using RT-PCR detection of flavivirus genus alphavirus genus, Bunyan virus, viral nucleic acid, the results 5 specimens yellow virus nucleic acid positive. The positive specimens No.: JB114, JB135 DLN24 RLB61 and RLC31 Determined after further nucleic acid detection and sequence analysis of five specimens, three (JB114, JB135, DLN24 virus strain) for the Japanese encephalitis virus, genotype Ⅰ 2 (JB114, JB135), a gene type Ⅲ (DLN24); strains of dengue virus on (RLB61 RLC31 virus strain), dengue type 1 1 (RLB61), Dengue 3 1 (RLC31). Molecular epidemiological analysis detected three Japanese encephalitis virus and dengue virus 2. Japanese encephalitis virus JB114 and JB135 strains of viral encephalitis patients from Jinghong City, cerebrospinal fluid, JE virus DLN24 strains from Dali City in cerebrospinal fluid of patients with viral encephalitis, three cases of Japanese encephalitis patients are local infections. The dengue virus RLB61 and RLC31 are from Ruili City, Dehong, unexplained fever patients with acute phase serum, two cases are imported cases of dengue fever patients, including one case for entry of Myanmar's Muse border residents care patients, other patients Ruili City Myanmar Travel border residents to return after the onset. From the molecular level for the first time the presence of dengue fever imported cases confirmed in western Yunnan border areas, border areas in Myanmar in 2008 and confirmed the existence of the prevalence of dengue type 1 and type 3 virus. The 106 specimens does not detect the alphavirus genus, Bunyan virus nucleic acid positive samples. Using real time PCR to Banna virus nucleic acid detection methods of these specimens, the viral nucleic acid detection publishing satisfied weakly positive specimens were the (MNF40, and MNF41) Ct values ??are 32. But ordinary RT-PCR method to detect the the Banna virus nucleic acid, negative were not MNF40 and MNF41. 4 Conclusion Yunnan cases of viral encephalitis, of genotype Ⅰ and Ⅲ of Japanese encephalitis virus infection cases, which strain of the virus from patients Xishuangbanna for gene Ⅲ type genotype Ⅰ Dali. Japanese encephalitis virus nucleotide and amino acid homology and phylogenetic analysis showed that the nature of Japanese encephalitis virus has better genetic stability. The western Yunnan border areas the presence of imported cases of Dengue type Ⅰ and type Ⅲ infection indirect proof of the presence of Ⅰ and Ⅲ dengue virus of the popular 2008 summer and autumn neighboring Myanmar border areas of the province at the same time. 2 dengue virus detected in Southeast Asia epidemic strains of nucleotide and amino acid homology and phylogenetic analysis showed that the closer the genetic relationship between them. Yunnan border area dengue prevention and control work should be strengthened in the future to prevent the local epidemic caused due to imported cases. Yunnan Province, in cases of viral encephalitis may Banna virus infection cases exist, should further carry out the investigation on the relationship of Banna virus and disease.
