Abstract

About 5 million hectares of saline soil, which salted 100 million mu of arable land, there are 300 million acres of salted wasteland to be of reclamation activities. Soil salinization important issues affecting agricultural production and ecological environment, and the cultivation of salt-tolerant plants, salt-tolerant mechanism of salt tolerance gene positioning separated by more and more people are concerned about. In this experiment, \F 2 groups as research material. SSR technology (BSA) method to locate the salt tolerance gene. 246 pairs of primers were screened, of which only 10 pairs between the two materials, polymorphism, the polymorphism index was 4.07%, between the two materials we used to hybridize the genetic background is very similar to the use of multiple pairs of primers the difference obtained by screening with a higher reliability. Only 10 pairs of primers WMS299 different salt-tolerant the Pool and sensitive pool. WMS299 F 2 groups of 80 individuals were amplified and leaves people a part of the salt-tolerant strains with salt-tolerant parents with type most sensitive monoclonal sensitive parents with type mutation sites showed that the WMS299 and salt-tolerant mutant chain relationship, and the sites are located in the long arm of chromosome 3B. To utilize Jionmap1.4 software mapping analysis, the WMS299 the locus genetic distance 14.003cM. Further separation of the salt-tolerant gene-based materials may lay the foundation. Laboratory reciprocal cross RH8706-49 with another salt-tolerant materials found F 1 each growth stage tolerance index higher than that of salt-tolerant pro RH8706-49 for the mother The combination of F 1 tolerance index than anti-crosses, this phenomenon shows that the salt tolerance of genetic may to some extent related to the plasmon. The second part of this experiment, RH8706-49, H8706-34, and their female parent Pu agricultural 3665 using RAPD method to study the differences in their mitochondrial DNA. Randomly selected 53 random primers, two primers RH8706-49-specific amplification products specific band recovery, cloning, sequencing analysis. And wherein one of the design of specific primers for specific amplification.
