Study on Hypoglycemic Effects and Mechanisms of Action of Columbin in Type 2 Diabetic Rats

Abstract


Objective: To establish a rat model of type 2 diabetes, study coffee and its effect on Currumbin hypoglycemic glucose transporter -4 (GLUT-4) protein expression and insulin receptor binding capacity, and to explore the possible drop coffee Currumbin Sugar mechanism. Methods: (1) high-sugar high fat diet three months combined with low dose streptozotocin (STZ) 30mg/kg intraperitoneal injection to establish experimental rat model of type 2 diabetes, animals were divided into five groups: normal control group, model group, low dose group Currumbin coffee (25mg/kg), coffee Currumbin high dose group (50mg/kg) and positive control group (metformin, 200mg/kg). Gavage 4W, 1 time / day, a fasting blood glucose (FBG), fasting serum insulin (FINS), insulin resistance index (ISI), triglyceride (TG), total cholesterol (TC), free fatty acid (FFA) , rat body weight, glucose tolerance was observed as the detection index hypoglycemic effect of drugs. (2) After 4 weeks, the rats were decapitated, some were taken right leg quadriceps, liver, skeletal muscle cell membrane and liver cell membrane extract total protein. Western Blotting assay using skeletal muscle glucose transporter (GLUT) -4 protein expression, radioligand receptor binding assay hepatic insulin receptor binding capacity. Results: (1) by the high fat diet, the rats fasting insulin (FINS), total cholesterol (TC), triglyceride (TG), free fatty acid (FFA) was significantly higher than the control group (p lt; 0.05) , insulin resistance index was significantly lower (p lt; 0.05). After administration of each dose group decreased body weight, no significant difference between the two groups (p gt; 0.05). During the experiment, the model control group blood sugar stable, to administration for 14 days, coffee Currumbin low dose group and high dose group, metformin group glucose values ??were compared with the model group decreased 15.04%, 19.65%, 20.18% (p lt; 0.05); until the end of 28 days of administration, blood glucose levels in each group were down 24.97%, 30.85%, 42.79%, and the model group, low dose group coffee Currumbin difference was significant (p lt; 0.05), coffee Lun Bin high-dose group and metformin group difference was highly significant (p lt; 0.01). Compared with the model group, drug treatment blood insulin levels did not change significantly (p gt; 0.05), while the model control group, insulin resistance index was significantly lower than the normal group (p lt; 0.01), after administration of insulin resistance index was significantly increased (p lt; 0.05). Compared with the model group, low-dose coffee Currumbin, Currumbin coffee high doses significantly improved glucose tolerance and metformin group (p lt; 0.05), blood glucose levels in each group at 0.5h, 1h, 2h were significantly lower (p lt ; 0.05), blood glucose values ??were within 0.5h after intragastric glucose peaked. Animals treated group TC, TG were significantly reduced compared with the control group, the difference was highly significant (p lt; 0.01), coffee Currumbin low and high dose group and metformin group free fatty acids, compared with the model group, difference was statistically significant (p lt; 0.01 or p lt; 0.05). (2) the model control group rat skeletal muscle GLUT4 protein expression was significantly lower than the control group (p lt; 0.01), after administration of coffee Currumbin low and high dose group and metformin group GLUT4 protein expression was significantly increased (p lt ; 0.01). (3) compared with the control group, type 2 diabetic model rats liver cell membrane insulin receptor dissociation constant KD was significantly higher (p lt; 0.01), receptor maximum binding capacity Bmax increased (p lt; 0.05). Each group after administration of insulin receptors in rat liver membranes dissociation constant KD significantly reduced (p lt; 0.01), Bmax decreased compared with the model group, coffee Currumbin low and high dose group and metformin group difference was significant ( p lt; 0.05 or p lt; 0.01). Conclusions: (1) coffee Currumbin experimental type 2 diabetic rats with hypoglycemic effect; (2) coffee Currumbin diabetic rat skeletal muscle cell membrane increases GLUT4 protein expression; (3) coffee Currumbin increased in diabetic rats liver cell membrane insulin receptor binding capacity.
