Abstract


Objective: To induced by TNBS (trinitrobenzene sulfonic acid) enema form of inflammatory bowel disease (IBD) model, through the detection level of myeloperoxidase (MPO) and the determination of of NF-KBp65 and p50 expression, explore Pulsatilla Decoction efficacy on rat inflammatory bowel acute phase mechanism. Methods: 40 male Wistar rats were randomly divided into normal control group, model group, the Pulsatilla low dose group, the Pulsatilla high dose group, mesalazine group, 75mg/kg dose enema modeling after 24 hours, in addition to the normal control group Pulsatilla Decoction and model group were given 0.9% saline, were given low dose, high dose and positive control drug, each group was given a daily average of 1 times of 14 days, while rats diarrhea, blood in the stool and record the weight change The experimental animals were sacrificed 16 days after modeling and drawn. Determination of the femoral artery after the end of treatment, blood serum C-reactive protein (CPR) and colon tissue MPO levels, and for immunohistochemistry, immunoblotting (Western Blotting), PCR observed NF-KBp65, p50 protein and its gene expressed. Results: Compared with the normal control group, the other groups of rat colon mucosa gross and histological scores and MPO activity were higher (P lt; 0.05), compared with model group, the gross and histological scores of treated rats colonic mucosa and MPO activity were lower (P lt; 0.05). The serum levels of C-reactive protein was not statistically significant. The results show of model groups colon tissue of NF-KBp65 the protein expression is higher than the normal group, and NF-KBp50 lower than the normal group (P lt; 0.05); Chinese medicine low dose group, high dose group of protein expression of NF-KBp65 lower than the model group Chinese medicine low dose group, high dose group of NF-KBp50 of protein expression were higher than the model group, while positive the NF-κBp50 of the control group is lower than the model group (P lt; 0.05). Colon tissue of the model group of NF-ΚBp65 the mRNA level higher than the normal control group (P lt; 0.01), while the NF-ΚBp50 mRNA level lower than the normal control group (P lt; 0.05); Chinese medicine high dose group and low dose group and the positive control group was lower than the model group, the Pulsatilla the soup high dose group, low-dose group of NF-ΚBP50 the expression is higher than the model group, positive control group, the mRNA levels of NF-ΚBp50 lower than the model group, Chinese medicine, the high-dose group and traditional Chinese medicine low dose group (P lt; 0.05). Conclusion: Pulsatilla the soup high dose group can reduce the NF-ΚBp65 its mRNA expression while enhancing of NF-ΚBp50 its mRNA expression may be one of its efficacy, but may vary and 5 - amino salicylic acid treatment mechanism.
