Abstract
Fuzi Lizhong Pills are important compound preparation in clinic,which have excellent efficacy on insufficiency of spleen-YANG,psychroalgia of gastric cavity,emesis,diarrhea and cold of extremity.In this paper,HPLC fingerprints were developed and used to control the quality of Fuzi Lizhong Pills,meanwhile the RP-HPLC method was also developed for the quantitative determination of glycyrrhizic acid and glycyrrhetinic acid in the preparation.1.The extraction method was optimized by orthogonal experiment.The index of chromatographic fingerprints(F)was used as an indicator to select extraction process.Thirty one co-possessing peaks were selected to establish the HPLC digitized fingerprints by taking 5-HMF peak as the referential peak.The fingerprints were evaluated by the software of "Digitized Evaluation System for Super-information Characteristics of the Traditional Chinese Medicine Fingerprints 3.0",and the "SPSS software" was used for the cluster analysis of the results.There were great differences on the chemical composition proportion and content of Fuzi Lizhong Pills from different batches.2.The method of dismantling drugs was applied to study the compatibility regularity of medicinal materials.The software of "Digitized Evaluation System for Super-information Characteristics of the Traditional Chinese Medicine Fingerprints 3.0" was adopted for the evaluation of the fingerprints.Under the chromatographic condition in this test,the content of chemical composition in principal drug Radix Aconiti Lateralis Preparata was rather low,Radix Glycytthizae offered greater contribution to the fingerprint,the interaction between Rhizoma Zingiberis and other medicinal materials was significant since Rhizoma Zingiberis can cut down the content of other composition.3.The mult-wavelength fusion technique was used in the study to develop a muir-wavelength fusion HPLC fingerprint of Fuzi Lizhong Pills.Copious information of the sample fingerprints were obtained quickly and accurately.The unified HPLC fingerprint and the relatively unified criterion of the characteristics for Fuzi Lizhong Pills were also developed,trying to achieve the standardized transform of the fingerprint.It provided a new method for the quality control of TCM.4.Preliminary study of rat serum fingerprints of Radix Aconiti Lateralis Preparata and Fuzi Lizhong Pills was carried out,and provided reference for the further research on active compound and mechanism of action for the preparation.5.Methods based on RP-HPLC/UV have been developed for the determination of glycyrrhizic acid and glycyrrhetinic acid in Fuzi Lizhong Pills.The samples manufactured in different factories were analyzed to determine the contents of the two compounds.The methods are both simple and accurate,and can be used for the quality control of Fuzi Lizhong Pills.
