Abstract


As people's living standards improve, more and more attention water safety issues, microbial contamination caused by the quality of health problems is also of concern. According to the World Health Organization estimates that each year hundreds of millions in the world of foodborne disease, 70% were due to consumption of a variety of pathogenic microbial contamination of food and drinking water caused. E. coli is the most important human and animal intestines and the largest number in a dwelling bacteria, most of the non-pathogenic, but when a number of pathogenic E. coli into the human body parts, can cause diarrhea, cholecystitis, cystitis. People infected with pathogenic E. coli symptoms of stomach pain, vomiting and fever, especially when the infected infants and the elderly, the consequences may even be fatal. E. coli as a widely used international water quality hygiene indicators commonly used to indicate the status of water contaminated by feces, thus its quality control to ensure health and safety. Therefore the establishment of a new method of rapid detection of E. coli on the environment and people's health improved quality of life is important. This paper studies outlined as follows: (a) by measuring the OD values ??and bacterial culture medium plate counts, the four major categories of 13 kinds of microbial growth promoting substances on the growth of E. coli, E. coli were screened out several pro- growth effect of a good material, so you can quickly detect E. coli culture medium for the future improvement of formulations provide experimental and theoretical basis. (2) in E. coli based on the chromogenic medium be improved, through the medium of E. coli chromogenic adding different concentrations of sucrose, potassium dihydrogen phosphate and Tween -20, improved chromogenic culture of E. coli based detection rate. After using a modified chromogenic medium for different E. coli E. coli were detected in water with GB multi-tube fermentation method to do a comparison, the detection rate than GB average of multiple tube fermentation method of 11.2%. (3) Application Tanon colony counting system, automated count of E. coli, thereby reducing the counting time. The use of the counting system for urban sewage, Suzhou River, mirror on the lake of E. coli were automatically counted and compared with manual counting, the method with the traditional method of manual counting results are basically the same, but counting speed, high degree of automation with a wide range of applications. (4) based on the exponential growth of microorganisms during the growing time and the real relations between the concentration of microorganisms, a growth curve was established spectrophotometric new method for rapid detection of E. coli, for the rapid detection of E. coli in the future research provides new ideas.
