Anti-gout Capsule Down-regulation of MMP-1 and TNF-α Proteins Expression in Acute Gouty Arthritis Articular Cartilage of Rats

Abstract


Objective:To observe the effects of anti-gout capsule on expression of the MMP-1 and TNF-a in articular cartilage tissues of acute gouty arthritis rats.Method:The fifty male SD rates were randomized to the control, model, positive control(cochicine 0.8mg/kg), and anti-gout capsule low dose(0.3g/kg), high dose group(1.2g/kg). Besides control group, the gout model was made in other groups by injected sodium urate into the joint cavity. Anti-gout capsule groups were given once a time for 5 days. Positive control and model group were given equal volume of water. The structural changes were observed after hematoxylin-eosin staining (HE) in articular cartilage of rats. The expressions of MMP-1 and TNF-a proteins were detected in articular cartilage tissues in rats with immuno-histochemistry staimimg.Results:(1) According to HE staining, obvious edema and large inflammatory cells exist in cartilage and marrow chamber in model group, compared with groupa. But, edema is gentler and inflammatory cells fewer in anti- gout capsule low doses and high doses and positive group. (2)As a result of immunohistochemistry, expression of MMP1、TNF-a is significantly increased in model group than groupa in Cytoplasm or nucleus (P<0.01). (3) Expression of TNF-a has no significant difference between colchicine group and model group(P>0.05), while MMP-1 is obviously down-expression(P<0.01).Conclusion:Anti-gout capsule can relieve cartilage damage in acute gouty arthritis rats, and down-regulate the MMP-1 and TNF-a expression in cartilage tissues. It suggests that the effect of Anti-gout capsule may related with the down-expression of MMP-1 and TNF-a in acute gouty arthritis articular cartilage rats.
