Abstract


Ceftriaxone Sodium (CS) is a kind of antibiotic medicament. Compared with penicillin, the possibility of shock caused by CS is lower; therefore, it is safe and popular in clinic. Since 1996, CS has a significantly development in our country. It is the No. 1 in the field of anti-infection medicines, and its preparation and materials have drawn our eyes in recent five years. So it is very important to determine the concentration of CS in medicament field and body fluid.Some quantitative analysis can be carried out by chemiluminescence analytical method that is based on the direct proportion between the concentrations of analyzable substance and its chemiluminescence intensity. Chemiluminescence determination is based on light photons, and has a advantage of such as high sensitivity. Flow injection analysis (FIA) technique is widely used in solution analysis and solution disposition based on its more advantages such as easy operation, simple apparatus and high automation. With the development and popularity of FIA, it will become a trend to use FIA instruments connected with other normal instruments. FIA has the advantage of quick analysis and high precision, and CL has the advantage of high sensibility. So the connection between them provides a new analytical method for trace elements determination.In this project, a new FIA—CL pipeline is used. It also shows the study on determination of CS by FIA-CL which is based on strong suppression effects on the luminol- K.3Fe(CN)6 chemiluminescence system under the condition of alkalinity. We study optimum condition of determination of CS using FIA-CL with satisfactory results.1. Using water boiler under the condition of constant temperature to heat CS solution to accelerate reaction speed:CS can inhibit the emission intensity of the luminol- K3Fe(CN)6 chemiluminescence system in basic medium. In room temperature, emission intensity of the luminol- K3Fe(CN)s chemiluminescence system enhance with increasing of mixing time, when CS is mixed with NaOH, emission intensity can not reach maximum in a long time, which shows that mixing time of CS and NaOH can influence emission intensity directly. So we use water boiler under the condition of constant temperature to reduce mixing time to reach maximal intensity.2. The application of the mixture of ice and water to regulate reaction temperature :After CS is taken out of the water boiler, the temperature has great effect on the luminescence values of the luminol- K3Fe(CN)6 chemiluminescence system. So the mixture of ice and water is used to control the temperature. As a result, the reproducibility of chemiluminescence reaction is improved.3.
