Objective: To investigate the chronic hepatitis B patients with hepatocellular carcinoma myeloid dendritic cell load of HBV antigen phenotype and functional changes . Methods: blood components separator collected 31 cases of chronic hepatitis B patients with hepatocellular carcinoma in peripheral blood mononuclear cells ( PBMC ) in vitro induced GM-CSF and IL-4 under the action of MDC differentiation , development, adding \(IL-1β, IL-6, TNF-α and PGE of 2 < / sub > ) to promote the mDC mature , and in promoting maturation factor respectively with HBcAg and HBsAg sensitization , mDC , LPS as control antigen , antigen detection sensitized on the the the mDC phenotype and function of the impact . Results: In chronic hepatitis B patients with hepatocellular carcinoma : 1.mDC surface molecule expression : immature MDC surface molecules CD80 , CD83 expression was significantly lower than the percentage of positive cells mature mDC (CD80: 9.3% ± 3.3% vs 34.0% ± 20.7 %, CD83: 4.5% ± 2.7% vs13.2% ± 6.5%, both P lt ; 0.05 ) ; by the the HBcAg sensitized mDC surface molecules CD80 , CD86 , CD40 , HLA - DR expression percentage of positive cells compared with antigen - induced Min significantly increased ( CD80 : 58.3 % ± 21.1 % vs 34.0 % ± 20.7 % , CD86 : 79.8% ± 12.9 % vs 45.2 % ± 22.0 % , CD40 : 69.8 % ± 12.8 % vs54.2 % ± 23.2% , HLA - DR: 78.4% ± 11.7% vs 54.6% ± 20.7%, both P lt ; 0.05 ) ; percentage of positive cells the HBsAg sensitized mDC surface molecule CD86 , HLA - DR expression compared with antigen ( CD86 significantly increased : 70.2% ± 9.1% vs 45.2% ± 22.0%, HLA-DR: 72.3% ± 3.9% vs 54.6% ± 20.7%, both P lt ; 0.05 ) ; sensitized by LPS mDC surface molecules CD80 , CD86 , CD40 , CD83 HLA - DR expression percentage of positive cells than those without antigen sensitization significantly increased (CD80: 57.0% ± 7.1% vs34.0% ± 20.7%, CD86: 96.5% ± 2.1% vs 45.2% ± 22.0%, CD40: 79.5 % ± 24.8% vs54.2% ± 23.2%, CD83: 46.5% ± 0.7% vs 13.26% ± 6.52%, HLA-DR: 97.5% ± 2.1% bvs54.6% ± 20.7%, both P lt ; 0.05 ) Cytokines secreted by 2.mDC : the HBcAg sensitized MDC secretion of cytokines IL-10 and IL-12 amount was significantly higher than non - antigen sensitized group (IL-10: 49.4 ± 33.6pg/ml vs18.6 ± 5.5 pg / ml, IL-12: 658.1 ± 306.7pg/ml vs 122.4 ± 87.1pg/ml, both P lt ; 0.05 ) ; HBsAg
