Abstract

Objective: To observe the antisense C-myc oligodeoxynucleotide (antisense c-myc oligonucleotides ASODN) joint 188 Re radiation therapy on intimal hyperplasia in rabbit iliac artery injury discussed ASODN 188 joint , Re radiotherapy prevention of restenosis potential role. Research Methods: To establish a rabbit iliac artery balloon injury model, the first use of a balloon catheter injury in rabbits iliac artery, and then 188 Re fluid through the balloon catheter, the rabbit iliac artery injury blood vessels after irradiation, the ASODN by pluronic gel drug delivery system and Lipofectin transfection import system, its role in the adventitia of the vascular injury, and 3 weeks after the vessel segments taken tissue sections and hematoxylin - eosin staining morphological observation under the microscope and by immunohistochemical staining of α-actin, and subjected to image analysis using an image analysis system, the intimal area and in the membrane area and the intima / media (I / M) ratio of the area is calculated, and the arterial endometrial α-actin staining positive cells percentage, and analyzed statistically. Results: 1. Vascular morphological changes: rabbit iliac arteries after balloon injury three weeks appears obvious stenosis, neointimal growth, proliferation of smooth muscle layer. Membrane area of ??the lumen, intima, in effect 2.ASODN: in ASODN intervention group surgery side with the control group operated side compared to the iliac arteries, increased lumen area (0.91 ± 0.23 mm 2 VS 0.72 ± 0.19 mm 2 , P (0.01), reduced neointimal area (0.18 ± 0.12 mm 2 vs 0.62 ± 0.18 mm 2 , P lt; 0.01), I / M area decreased (0.344 ± 0.05 VS1.02 ± 0.15, P lt; 0.01) 3 188 Re irradiation lumen, intima, medial area influence: 188 Re irradiated group 8Gy dose of the operated side compared with the control group operated side iliac artery lumen area was significantly increased (0.72 ± 0.18 mm 2 vs 0.724 ± 0.19 mm 2 , P gt; 0.05), neointimal area significantly reduce (0.52 0 .17 mm 2 vs 0.62 0.18 mm 2 ,, P gt; 0.05), the I / M area than non-significantly decreased (0.92 ± 0.09 VS 1.02 0.15, P gt; O.05). 15Gy dose surgery side compared with the control group operated side iliac artery lumen area increased (0.90 ± 0.22 mm 2 vs 0.72 ± 0.19 mm 2 ., P lt; 0.01), reduced neointimal area (0.25 0.13 mm vs 0.62 0.18mm 2 P lt; 0.01), the I / M area decreased (0.42 ± 0.09 VS 1.02 ± 0.15, P lt; 0.01) 188 4.ASODN joint Re irradiation of the lumen, intimal medial area Effect: (1) of ASODN joint 188 Re irradiation (15Gy) surgery side iliac artery compared with the surgical side of the control group increased lumen area (0.99 ± 0.21 mm 2 vs 0.72 ± 0.19 mm 2 , P lt; 0.01), and reduced neointimal area (0.04 ± 0.015 mm 2 vs 0.62 ± 0.18 mm 2 P lt; 0.01), the I / M area ratio decreased (0.08 ± 0.018 VS 1.02 ± 0.15, P lt; 0.01). this group ASODN lumen area no significant increase (0.99 0.21 mm 2 the vs and 0.91 ± 0.23 mm 2 , P gt; 0.05), reduced neointimal area (0.04 ±
