Abstract


In March 2003 , a new coronavirus (CoV)-SARS coronavirus is recognized pathogen causing SARS (severeacute respiratory syndrome) . In the present study , we have obtained 10 direct source of SARS patient stool or throat swab samples of their cells subcultured complete genome sequence of the SARS-CoV to , and in conjunction with the 115 SARS-CoV genome sequences in public databases system compare analysis . GZ02 sequence as a reference, found 266 exist simultaneously in two or more genomic single nucleotide polymorphism (SNP) locus . These polymorphic loci in the genome of the SARS-CoV skewed distribution . Also found that 18 major missing type and two kinds of insert type defines two kinds of genotypes associated with the onset period and mortality . S protein by the onset of early positive selection pressure , and then gradually stabilized . We apply the ortho connection method (neighbor-joining) to construct phylogenetic tree of 125 SARS-CoV . By direct sampling from the same patient sample and its cells subculture of SARS-CoV genome sequence comparison analysis found that almost does not produce mutations in the genome sequence of the SARS-CoV in cell culture process , but will affect the sampling time strategy process. Speculated that this phenomenon may be the cells subcultured process will select somehow predominant strains of virus caused .
