Study of Inhibitory Effects of Antisense Oligonucleotides on b-Catenin c-myc VEGF Gene Expression in Human Hepatocellular Carcinoma Cell

Abstract


Objective: To study for beta - catenin , C - myc antisense nucleotide expression levels of VEGF , explore SMMC7721 hepatoma cell line expression of VEGF in C-myc gene regulation and for beta - catenin , C -myc antisense gene therapy in liver cancer gene therapy prospects . Methods: (1 ) the routine culture the liver cancer SMMC7721 cell lines and seeded in 6-well plates . ( 2) According to the the Genebank included beta- catenin , C - myc mRNA sequence design antisense nucleotides for translation initiation site . (3 ) translation initiation site for beta - catenin antisense the mismatch oligonucleotide translation initiation site for the C-myc antisense mismatch oligonucleotide -treated cells , and blank control group , cells were collected after 24 hours of incubation under hypoxic conditions to extract total RNA . (4) by RT-PCR and FR200 image analysis system semi-quantitative analysis of the relative expression of beta - catenin , C - myc and VEGF expression to beta - actin expression for each gene within the control . ( 5) Application of X2 test for statistical analysis data . Results: (1 ) for beta - catenin translation initiation site of the antisense oligonucleotide significantly inhibited the expression of beta - catenin mRNA , antisense group and missense group inhibition rates were 0.73 ± 0.06 and 0.10 ± 0.07 . VEGF expression by inhibition of VEGF inhibition rate was 0.80 ± 0.05 and 0.09 ± 0.06 . (2) inhibition of the expression of c - myc mRNA translation initiation site for the C-myc antisense oligonucleotides , antisense group and missense group inhibition rates were 0.75 ± 0.04 and 0.11 ± 0.05 , while VEGF expression was inhibited VEGF inhibition rates were : 0.60 ± 0.07 and 0.08 ± 0.03 . Conclusion: (1 ) According to the the Genebank included beta- catenin , c - myc mRNA
