Objective: To compare type 2 diabetic rats and normal rat kidney tissue differences in the gene expression profile of the gene chip technology to explore the pathogenesis of diabetic nephropathy (DN) . Methods: (1) the experimental rats were divided into two groups, a group of normal control group (10) , another group of small dose streptozotocin (STZ) fed a high fat diet for a type 2 diabetic rats group ( 12) ; (2 ) modeling during the monitoring body weight , blood glucose , blood lipids , serum insulin , parallel glucose tolerance test ; ( 3) , respectively, from the two groups of rats kidney tissue extracted mRNA, by reverse transcription with Cy3 and Cy5 fluorescent marker, to obtain cDNA probes of the two sets of animal origin ; (4) of the different sources of cDNA probes for hybridization with the gene fragment containing 4000 chips , and the results are scanned by the scanner and the software for statistical analysis ; (5) according to the chip the result of the selection of a number of differentially expressed genes , application of semi - quantitative RT-PCR validation . Results: ( 1 ) 12 weeks after the end of the model , type 2 diabetic rats compared with controls body weight was significantly increased, elevated blood lipids , fasting glucose and fasting insulin levels , abnormal glucose tolerance test was significantly decreased insulin sensitivity index ; ( 2) of the 4000 genes screened a total of 399 differentially expressed genes in the two groups of rat kidney tissue , which dropped to 267 , up 132 ; promote lipolysis in type 2 diabetic rat kidney gene , oxidative bowel -related apoptosis-related insulin secretion associated mesangial extracellular matrix synthesis and gene expression coagulation raised ; promote glucose utilization , energy metabolism , antioxidant related mesangial extracellular matrix degradation fibrinolytic related insulin signal transduction genes were downregulated . (3) verify that the part of the gene expression microarray results by semi- quantitative RT-PCR method . Conclusions : ( 1) the experiment successfully constructed experimental type 2 diabetes model , obesity, hyperglycemia , hypertriglyceridemia , insulin resistance , type 2 diabetes characteristics , is the study of the ideal of type 2 diabetes and its complications animal models. (2) material energy metabolism , oxidative stress enhanced coagulation and fibrinolysis system dysfunction, enhanced apoptosis is closely related to the pathogenesis of type 2 diabetic nephropathy .
