The Role of SIP1 and Smad4 Protein in Tumor Invasion and Metastasis of Hepatocellular Carcinoma

Abstract


[Objective] To explore the the SIP1, Smad4 protein , TGF - β1 and epithelial cadherin (E-Cadherin) in hepatocellular carcinoma (HCC) invasion and metastasis process mechanism . [Methods] In situ hybridization of Smad4 , TGF-β1, E-Cadherin expression of SIP1 and immunohistochemical methods in 60 patients with HCC specimens studied to observe the relationship between these four indicators with HCC invasion and metastasis - related indicators . Select cirrhosis specimens of 15 cases of surgery for the same period , 15 cases of normal liver tissue as a control group . Statistical Methods SPSS11.5 statistical package . [Results] 1 . Cancerous tissue and cancerous liver tissues SIP1 the positive expression rates were 60.0% ( 36 / 60 ) and 35.0 % (21 /60 ) ; expression of Smad4 protein positive expression rate was 65.0% ( 39 / 60 ) and 31.7% ( 19/ 60) ; positive expression of TGF - β1 in 68.3 % ( 41 / 60 ) and 41.7% ( 25/ 60) ; expression of E -cadherin to reduce the rate of 71.7% ( 43 / 60 ) . The 2.SIP1 expression in HCC tumor size , pathological grade and whether portal vein tumor thrombus was significantly positively correlated (P ≤ 0.05), with encapsulated or not was a significant negative correlation ( P ≤ 0.05 ) ; Smad4 protein expression encapsulated showed a significant negative correlation ( P ≤ 0.05 ) ; TGF- β1 expression in HCC tumor size and number of lesions was a significant positive correlation ( P ≤ 0.05 ) ; expression of E -cadherin with portal vein tumor thrombus was highly significant negative correlation ( P ≤ 0.01 ) . 3.HCC in SIP1 of expression and of TGF - β1 of expression was significantly positive related ( P ≤ 0.05 ) , with the expression of E-cadherin was significant with negative related ( P ≤ 0.05 ) ; of Smad4 protein 's expression and of TGF - β1 of expression was significant with positive correlation (P ≤ 0.05), and E-cadherin expression was significantly negatively correlated ( P ≤ 0.05 ) . [Conclusion] 1.SIP1, TGF-β1 can promote the invasion and metastasis of HCC , the loss of expression of E -Cadherin promotes the invasion and metastasis of HCC the Smad4 protein involved in the invasion and metastasis of HCC , the specific role of controversial . 2.HCC in SIP1 , TGF - β1 , E- Cadherin as indicators reflect HCC invasion and metastasis characteristics . 3 . TGF-β1/Smad ways to inhibit or directly inhibit the expression of E-Cadherin is to the SIP1 promote one of the mechanisms of HCC invasion and metastasis .
