Abstract


[Objective]: through the establishment of cochlear ischemia-reperfusion animal model: 1. Observe cochlear ischemia and reperfusion in different time periods morphological changes 2. Observe normal cochlea isomer distribution of nitric oxide synthase 3 pairs ischemia and reperfusion in different time periods cochlear tissue distribution of nitric oxide synthase isoforms image analysis 4. study of dexamethasone in cochlear ischemia-reperfusion injury in rats [method]: healthy guinea pigs were randomly divided into normal group, sham operation group, ischemia group, 30 minutes, 6 hours reperfusion group and 24 hours reperfusion group. After four sub model group and dexamethasone treatment group. Cochlear ischemia-reperfusion model: First use of micro-artery clip side of the common carotid artery occlusion and bilateral vertebral artery cauterization establish cochlear ischemia model, open the folder arterioles establish reperfusion model. Histochemical ischemia and reperfusion was observed at different time points of various parts of the cochlea histological changes, and with the use of dexamethasone to compare the results; using immunohistochemistry (ABC) method to detect the animal cochlea nitric oxide synthase isoforms and changes, and image analysis. [Result]: HE staining in normal group and sham operation group organ of Corti hair cells without injury, vascular pattern clear; ischemia and reperfusion in each group are visible inner and outer hair cell deformation, Corti is inner and outer hair cell defects, vascular pattern thinning. Dexamethasone group than in the corresponding cells reperfusion Zhu deformation, defects were reduced thickness slightly increased vascular pattern. Immunohistochemical observation and image analysis showed that: nitric oxide synthase isoforms in normal stria vascularis, spiral ganglion cells, Corti's inner and outer hair cells, vascular ball, spiral ligament, spiral limbus has weakly positive to expression. In each group, ischemia, nitric oxide synthase isoforms in the vascular pattern, spiral ganglion and the organ of Corti are present upregulated. Reperfusion groups exist only increased iNOS expression. Dexamethasone significantly inhibited 24 hours reperfusion group the expression of iNOS and eNOS accompanied by upregulation of expression. [Conclusion]: through the micro-artery cauterization able to create folders and cochlear ischemia-reperfusion animal model. Ischemia and reperfusion group at different times, such as pathological changes associated with inner and outer hair cell deformation, defects, vascular pattern thinning and so on. Nitric oxide synthase isoforms in normal cochlea has weakly positive to positive. iNOS may be involved in cochlear ischemia-reperfusion
