Abstract


Objective: To study the role of the aldosterone receptor antagonist spironolactone acute myocardial infarction myocardial apoptosis and apoptosis-related genes . Methods : by ligation of the left anterior descending artery caused by a large area of left ventricular myocardial infarction model . Postoperative survival of 48 SD rats were randomly divided into a control group of myocardial infarction (AMI group , n = 6 , saline 2ml / d orally ) and spironolactone treatment group (Spi group , n = 6 , spironolactone 20mg / Kg / d, 2 ml gavage) , another sham group (Sham group , n = 6 , and saline 2ml / d gavage) . After 48 hours , 7 days, 14 days , 21 days to observe the following indicators : the chloramine-T method for the determination of the collagen content of rat non- infarcted myocardium ; ② by immunohistochemistry method determination of non-infarcted myocardium Ⅰ / Ⅲ collagen ratio ; ③ non-infarcted myocardium, the rate of apoptosis and Bcl-2, Bax protein expression was determined using flow cytometry ; determination of non - infarcted area cardiac myocytes of Bcl-2mRNA and the BaxmRNA expression ( 4 ) by PCR . Results: 1 . Spironolactone non- infarct zone myocardial tissue collagen content : Sham group phase comparison , AMI group , Spi group of non-infarct zone myocardial tissue collagen content in 48 hours without significantly with the difference , 7 days , 14 Tian , 21 Tian are significantly higher (P lt; 0.01-0.05); compared with the AMI group , Spi group 48h no difference ( P gt ; 0.05 ) , 7 days and 14 days has been reduced reduced by 2% and 15% , respectively (both P gt ; 0.05), and day 21 decreased by 35% (P lt; 0.05). Spironolactone non-infarcted myocardium Ⅰ / type III collagen ratio : Sham group compared with the AMI group and Spi rats non-infarcted myocardium I / III collagen type than 48h when no difference ( P gt ; 0.05) , 7 days ( P LT ; 0.05 ) , 14 days and 21 days were significantly
