Association of Gene Mutations of Luteinizing Hormone β-subunit and Inhibin α-subunit with Premature Ovarian Failure

Abstract


Objective: Premature ovarian failure(POF) was defined as secondary ameno-rrhoea with elevated gonadotrophins occurring under the age of 40, which affects 1% of women in the general population. POF is a highly heterogeneous condition and the aetiology of it varied from different races and different districts. At the present time,some genes ,such as luteinizing hormone(LH) gene and inhibin(INH) gene,have been found to be associated with the susceptibility to POF. This study is to probe into the association of the Trp8Arg,Ile15Thr and Gly102Ser mutations in LH P-subunit gene and 769G-A mutation in INH α-subunit gene with POF in Hubei province in China.Method: A total of 124 unrelated women (34 case of POF and 90 controls) in Hubei province in China were screened for the mutations of the Trp8Arg, Ile15Thr and Gly102Ser in LHβ gene,and INHα gene 769G-A polymorphism was performed in 34 POF patients and 125 controls in Hubei province in China by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).Result: The frequencies of Arg8, Thr15 and Ser102 alleles in LH β gene in POF were significantly higher than that in control groups(x~2=12.192, 9.610, 5.327, P<0.05).The Arg/Arg and Thr/Thr genotype frequencies of POF in LH β gene were also significantly higher than that of control groups(P<0.05). There was no significantly difference in Ser/Ser genotype frequencies in LH β gene between POF and control groups(P>0.05). INHα gene 769G-A point mutation frequency in POF group was 5.88%,significantly higher than controls(Fisher’s exact,P<0.05).Conclusion: The Trp8Arg and Ile15Thr mutations in LHβ gene have correction with POF in Hubei province in China, and the Gly102Ser mutations did not have correction with POF in Hubei province in China. The 769G- A point mutation in INHα gene is associated with the happen of POF in Hubei province in China.
