Benzene is an extremely versatile industrial organic solvents, it is as important to the production of raw materials and organic solvent widely used in many areas of production, and long-term exposure to benzene can be caused by benzene poisoning, aplastic anemia and leukemia, but so far on benzene poisoning, benzene caused leukemia pathogenesis has not yet been fully elucidated. Recent years, with the development of molecular biology techniques, benzene caused leukemia mechanisms are from the level of genes to explore, but has yet to have the system about the benzene poisoning crowd genes differentially expressed. Objective: To study the tumor-related differential gene expression changes in peripheral blood leukocytes of varying degrees of benzene poisoning workers, to explore the molecular mechanisms of tumor exposure to benzene causes. Method: Select statutory diagnosis of occupational diseases diagnosed by 6 shoe operating benzene poisoning workers and a suspected the benzene poisoning workers as the experimental group, six benzene poisoning workers were chronic mild benzene poisoning two cases, moderate chronic benzene poisoning 2 patients with chronic severe benzene poisoning one cases of aplastic anemia in 1 case; benzene-exposed workers will not contact the poison as a control. Experimental and control groups in our follow-up survey three months had no history of medication, the study group and the control group by age (the difference is less than 3 years old), gender (all female), education level, smoking history, history of alcohol consumption etc. than pairing. Isolated from peripheral blood leukocytes leukapheresis solution to the Trizol extracted RNA, agarose gel electrophoresis of RNA quality, purified RNA, followed by reverse transcription hybridization probe labeling, the experimental group samples using reverse transcription Cy3 fluorescent marker of cDNA (was red), control group samples using reverse transcription Cy5 fluorescently labeled cDNA (green). Containing 2780 cDNA clone using seven microarray tumor associated gene chip hybridization, hybridization signals were scanned with a scanner, and then the image is converted to a digital signal based on the fluorescence intensity, the use of cluster analysis software, for screening out having a specific tumor-associated gene expression analysis. Experimental results: specific differential expression of 44 genes found in the tests of seven 2780 tumor-associated gene chip, These genes appear upward or downward trend. All genes are in GENEBANK login.
