This article is a sub-project of the major projects of the Ministry of Science and Technology 15 scientific and technological food safety issues. In this paper, the main problem of domestic fruit and vegetable pesticide residues detection system from two aspects: the formulation of the original commercialization acetylcholinesterase chromogenic reagent system optimized reagent cost; the subcritical water extraction technology used in the extraction of pesticide residues, and compared with several other pretreatment technologies. The main contents include: the acetylcholinesterase color reaction system detected pesticide residues is an ideal and reliable method, but the higher cost of existing reagent formulations detection. Ensure high sensitivity under the premise, a lower cost formulation was determined by comparing the study, i.e. the enzyme and the substrate used was changed to 3/4 of the original recipe, the coloring agent is used in an amount changed to 1/2 of the original formula The overall savings of 1/3 of the reagent costs. F test to prove after adjustment formula and the original recipe was no significant difference between the. The adjusted recipe the three pesticides quantitative detection limits were: methamidophos 0.3mg / L, the trichlorfon 0.04mg / L, the dichlorvos 0.02mg / L, and the National standard fast measurement method consistent. . Trichlorfon GC-ECD analysis conditions, the standard curve was linear correlation coefficient R = 0.998, measuring range 10-200μg / L. Comparative study directly oscillation extraction, ultrasonic extraction, subcritical water extraction and microwave-assisted extraction extraction of four pretreatment methods trichlorfon in the laboratory simulation cabbage samples. Gas chromatographic analysis results show that pesticide residues in cabbage trichlorfon, higher extraction rate of subcritical water extraction and microwave-assisted extraction technology than manually oscillation and ultrasonic extraction. Test of one kind of fruit and vegetable pesticide residues pretreatment technology - subcritical water extraction technology, acetylcholinesterase photometric detection analysis. Simulate Sample subcritical water cabbage in the the methamidophos pesticide residue extraction of optimal conditions for the temperature of 100 ~ 120 ℃, time 2 ~~ 4min. GB oscillation method and ultrasonic method, subcritical water extraction with high extraction rate, good precision and easy to operate. And subcritical water fast extraction device designed to carry out a preliminary study. The research results have been applied to the development of our group 'multi-channel monitor food safety' by the Hubei Provincial Science and Technology Department identified the technology reached the international advanced level.
