Objective: The pathological changes of cerebral ischemia injury microvascular permeability change as well as damage to the integrity of the blood vessels , leading to neuronal damage and pathophysiological changes , and led to a series of complications . Vascular integrity is guaranteed by the intimal , including anatomy and the integrity of the barrier function of the blood brain barrier (BBB) ??and the basement membrane . Brain microvascular endothelial cells form the structural basis of the blood - brain barrier (BBB) ??. BBB include a three-tier structure : cerebrovascular endothelial cells , the foot end of the basement membrane and star-shaped cells . Laminin (Laminin, LN) is a component constituting the base film structure , important role for the maintenance of vascular integrity , may reflect the damage of the basement membrane . Nerve cell injury in ischemic cerebral infarction and apoptosis - related apoptosis by Bax gene regulation . To further explore the the brain ischemic brain microvascular endothelial cell damage repair and apoptosis mechanisms , as well as defibrase treatment of ischemic cerebrovascular disease mechanism , we adopt a rabbit ischemic infarction model of the high fat and sugar , drop after the intervention defibrase monitoring the rabbit arterial plasma within Laminin expression changes in the brain tissue Laminin Bax expression , the observation defibrase high fat and sugar the rabbit cerebral infarction model Laminin concentration and its expression in brain tissue the impact , as well as the expression of apoptosis-related proteins in the brain tissue , and to explore the mechanism of the treatment of ischemic stroke defibrase . Methods : 24 New Zealand rabbits were randomly divided into non - high fat and sugar infarction model in the control group ( the control group) , high fat and sugar infarction model in the control group ( referred to as the model for the control group ) , drug intervention group ( or group) of defibrase . Except blank control group , to give a high-fat high - sugar diet , high fat and sugar model produced in rabbits , each group using the legal system writers rabbit infarction model through carotid artery microemboli autologous blood injection , and the drug intervention group ( drop defibrase group) intervention 7 days the defibrase (0.1U/kg) to dynamically detect arterial plasma lipids, blood glucose changes , enzyme immunoassay (ELISA) to detect Laminin changes . And common pathological observation the the detected brain Laminin and apoptosis regulatory protein Bax expression by immunohistochemistry (SABC) , calculating the integrated optical density and area of ??positive cells .
