Abstract


Objectives 1) To explore the expression patterns of Survivin, Bcl-2 and Bax in human medulloblastoma cell line UW228-2,-3. 2) To analyze the effects of resveratrol on the expressions of Survivin, Bcl-2 and Bax in human medulloblastoma cell line UW228-2,-3 with different chemical drugs resistant potentia 3) To study the status of Survivin and Bcl-2 expression in human medulloblastoma cells in vivo. Through Investigation further, 1)To elucidate the molecular mechanism of apoptosis in human medulloblastoma cells induced by resveratrol;2) To elucidate the expression patterns of Survivin,Bcl-2 and Bax in two different medulloblastoma cell lines after the treatment of resveratrol and to investigate the effect of resveratrol mediated cell growth inhibition,cell differentiation and apoptosis in in two human medulloblastoma cell lines(UW228-2,-3). To expect to know thoroughly the potential action mechanism of resveratrol on medulloblastoma cell line. To provide more laboratory evidence and theoretic basis for better treatment of human medulloblastoma.Methods Human medulloblastoma cell line UW228-2,-3 was cultured in DMEM (Dulbecco’s Modified Eagle’s Medium) culture medium containing 10% Fetal Bovine Serum. The cells were treated with resveratrol (100μM) ,when cells were in good state, then observe the effects of resveratrol-induced differentiation and apoptosis of medulloblastoma cells. Putting sterilized cover slides in the dishes before cell seeding whenever cell shape observation or immunocytochemical analysis was performed. After above treatments, cover slides were collected and fixed with 100% cold acetone. The cell proliferation situations of experimental groups were examined through Typran Blue counting method. Immunocytochemistry (ICC), Western-blot hybridization and RT-PCR were used to analyze the expression levels of Survivin, Bcl-2 and Bax., and the correlations of Survivin, Bcl-2 and Bax expressions with cell differentiation and
