Abstract


Since the eighty years latter of twenty century, Vacanti and others made use of tissue engineering to rehabilitate bone tissue disfigurement. Bone tissue engineering has been developed greatly and regarded as the most promising and feasible science of reconstruction in the future. The porous scaffold plays a key role in tissue engineering because of promoting cell attachment, proliferation, differentiation, and providing pathway for biofluids.Tricalcium phosphate(TCP) has been demonstrated with good bone conductivity, biocompatibility and long-term reliability. It has played an important role in surgical and dental implants. However, porousβ-TCP cell scaffolds do not meet the requirements of bone tissue engineering for lacking of strength and interconnection. Improving the intensity and interconnection of porousβ-TCP scaffold has become the focus in bone tissue engineering.In order to overcome the disadvantages ofβ-TCP synthesis with traditional technics, a newly synthesized technics called subsection liquid phase reaction was put forward, and potentialβ-TCP for fabrication scaffold was prepared after probing into feasible experimental conditions. Secondly, a simple spray device used for production spherical porogen was designed and fabricated according to the principle of spray. The self-made device possessed high efficiency when spherical porogens of stearic acid and paraffin were sprayed. Porogens made of stearic acid and paraffin were parts of the basic materials for porousβ-TCP scaffold formation. Their properties, such as configuration, structure, and diameter had effect on the scaffold’s characters. In the technique of production of porousβ-TCP scaffold, a new technics called porogen enwrap method was set forward for the first time. The homogenous mixture of porogen andβ-TCP powders were formed, and interconnected porousβ-TCP scaffold was finally fabricated. To increase the strength of interconnected porousβ-TCP scaffold, an additive (K2HPO4) was chosen to improve sintering process, and the effect of the additive concentration on strength was studied.The characters ofβ-TCP cell scaffolds, such as composition, granularity, density, micro-structure, porosity, water-up- take and so on were analyzed with IR, XRD, TEM, SEM and some chemistry methods. The feasible method for preparation high strength and interconnectionβ-TCP cell scaffolds was founded, and might offer a reference for others.
