Tissue Engineering is a science based on fundamentals, theories, techniques and methods of engineering and life sciences for construction bio-active implant system to replace or restore mal-functional tissue or organ. Tissue Engineering not only offers a new therapy method for relieving patient, but also provides new thought of copying the“tissue”and“organ”. It indicates that an epoch of bio-science and technology human body is coming, and new era of regeneration medicinal is coming. It’s a far-reaching signification revolution. Tissue Engineering involves three key factors, that is seeding cell, cell scaffold and tissue construction. Cell scaffold affects cell bio-action and foster efficiency, and decides cell’s adaptation, combination and rehabilitation after implantation. Cell scaffold is a hinge factor restricted tissue engineering clinical application. It is very important to manufacture cell scaffold with properties, for instance, good biocompatability high porosity and interconnection, to meet the requirements of tissue engineering.Sodium chlorate spherical porogen with variable size was prepared by solution dispersion technique. A modified Solvent Casting / Particulate Leaching, called Porogen Bonding-Solvent Injecting/Particulate Leaching, was developed to build PDLLA cell scaffold, the effect of experimental conditions on pore structure, configuration and interconnection was studied. In order to improve hydrophilicity and remain inner-structure of PDLLA scaffold, gelatin hybrid was employed to scaffold surface modification, and surface pretreatment, gelatin attachment and cross-linking were discussed in detail. Degradation of scaffolds in vitro were characterized, the factors related were discussed. A method for detection pH value of micro-liter solution was established to assay acidity of inner part of scaffold as degradation taking place.Column PDLLA scaffolds of 0.818±0.029cm in diameter and 0.260±0.028cm in height were fabricated, the porosity of the porous PDLLA scaffold was over 95%. It was showed that the scaffold was homogeneous in pore distribution, regular in configuration, fully interconnected between pores, suggested that the Porogen Bounding-Solvent Injecting/Particulate Leaching was the fast and useful method to fabricate PDLLA scaffold satisfied Tissue Engineering requirements. Water uptake of gelatin hybridized scaffolds was 1944.90±334.88%, which was almost twice larger than that (1198.87±374.77%) of unmodified scaffold, indicated the hydrophilicity of scaffolds
