Effect of Different Pathways and Doses of Inoculation on Establishment of Experimental Tuberculosis in SD Rats

Objective: To establish a of SD rats tuberculosis model (including the route of inoculation and the amount of inoculum ) , comparing different routes of infection and the amount of inoculum role in the establishment of the SD rat animal model of tuberculosis . Subjects and Methods: 70 SD rats were randomly divided into normal control group (A ) tail vein the 0.1mg infection group (B ) , the tail vein 1mg infected group (C ) , the tail vein 10mg infected group ( D group ) , intra-abdominal of 0.1mg infection group (E ) , intra-abdominal a 1mg infection group (F ) and the abdominal cavity 10mg infected group (G group ) , 10 in each group . Standard human strains of TB H 37 Rv0.1mg , 1mg 10mg three inoculant respectively injected via the tail vein and intraperitoneal two ways SD rats . At the same time , the control group were injected with the same volume of saline as a control. Were killed and six weeks after rats were weighed and calculated comparing lung , organ mass index of liver, spleen , lung , liver , spleen general lesions and microscopic characteristics , lung , liver, and spleen tissue homogenates smear done acid-fast staining and homogenate cultures observe whether mycobacterial colony growth . Results: 1 . Tail vein 1mg infection group , lung weight index , the tail vein 10mg infection group and the normal control group was significantly increased ( P <0.05 ) . The tail vein 10mg infection group , intraperitoneal 10mg infections in liver weight index was significantly higher than the control group ( P <0.05 ) . The tail vein of 1mg infection group , the tail vein 10mg infection group , abdominal 10mg infection spleen weight index and the normal control group significantly increased ( P <0.05 ) . 2. The tail vein 10mg infected lung , liver , spleen weight index with celiac 10mg infection group lung , liver , spleen weight index , there were significantly increased ( P <0.05 ) . 3. HE staining of lung , liver , spleen , see TB nodules varying degrees -added performance that lesions epithelioid cells and macrophages gathered into a group , surrounded by lymphocytic infiltration . With the increase in the inoculum size , the pathological changes of the organs and tissues in the more significant . Inoculant consistent tail vein infection in lung , liver , spleen pathological changes significantly more abdominal infection .
