Abstract


The first part : the middle cerebral artery ischemia reperfusion model purpose : to establish a simple, reliable , and less invasive Sprague-Dawley rats with focal cerebral ischemia model . Method: the suture method to reference Zealonga and Koizum , to improve production of Sprague-Dawley rats with focal cerebral ischemia model , and subsequently underwent neurobehavioral examination , TTC staining judge infarction results in order to observe the model reliability . Results: The model rats neurological dysfunction , brain tissue ischemia and infarction . Conclusion: The modified model prepared simple , reliable results , trauma , good stability, is the ideal experimental animal model used to study focal cerebral ischemia . Part II: cerebral ischemia reperfusion injury NGB SOD , MDA changes Objective: To study the rat cerebral ischemia-reperfusion injury cerebral myoglobin (neuroglobin, NGB) and serum superoxide dismutase ( superoxide dismutase , SOD expression changes ) , malondialdehyde (malondialdehyde, MDA) and reperfusion time relationship . Methods: The rat cerebral ischemia-reperfusion injury in animal models , different times of the cerebral cortex in the determination of injury ischemic penumbra the NGB change and serum SOD activity and MDA content changes , compared with sham-operated group . Results: Compared with the sham group at each time point , the cerebral cortex ischemic penumbra NGB positive neurons increased and then decreased the trend ; SOD activity first and then decreased , MDA content increased at first and then decreased , and two the existence of a negative correlation . Conclusion: cerebral ischemia and reperfusion injury NGB , SOD , MDA, changes in a certain time regularity , prompted NGB adaptive protective effect of ischemic brain injury , SOD and MDA serological level changes can be a better representative repair the damage situation of the nerve cells in the brain , may be a new quantitative indicators , play a role in the hypoxic-ischemic brain injury disease early discovery and treatment evaluation .
