Order to study the characteristics of the microorganism in the biological granulated fluidized bed sewage treatment reactor , respectively, from a biological granulated fluidized bed 10cm, 60cm, 110cm sampled at (water and particles of sludge), and the use of traditional cell culture method , detection of the bacteria The total number of denitrifying bacteria and anti - sulfide bacteria . Meanwhile, the application of molecular biology to study microbial characteristics in the bio- granulation fluidized bed . Granular Sludge bacterial genomic DNA directly extracted by cell lysis to the true bacterial and archaeal 16S rRNA gene universal primers 530F/1490R PCR amplification of genomic DNA of the the extracted bacteria in granular sludge , about 1kb PCR amplification products purified and separated by denaturing gradient gel electrophoresis (DGGE) for microbial community DNA fingerprint analysis , the characteristic bands for recycling sequencing and DGGE separation of the phylogenetic tree . Bacterial culture results show , the entire cross-section of the bio - granulation fluidized bed reactor , especially in the bottom of the reactor , there is a very rich aerobic microorganisms . Reactor also exist , such as denitrifying bacteria , anti- bacterial sulfide facultative anaerobic and obligate anaerobic microorganisms . PCR-DGGE results showed that the microbial community in granular sludge biological granulation fluidized bed reactor is very rich , in at 10cm microbial species reach 23 , 60cm at 21 and 110cm at 20 ; biological granulation fluidized bed at different heights has some endemic genera and Total species reactor at different heights microbial community succession is not obvious, the microbial community similarity of 83.1% , a more stable community structure . Phylogenetic analysis of bio- granulation fluidized bed microbial majority belong to the bacterial domain covers 18 taxa of bacteria domain , of which 61% ( 11 / 18 ) belong to the Proteobacteria , 17% ( 3 / 18 ) belong to the actinomycetes , 11% ( 2 / 18 ) of the low- GC Gram- positive bacteria , 11% ( 2 / 18 ) belong to other unknown bacteria .
