Effect of Swimming with Different Workload on the Gastrocnemius of the Endplates of the Mouse and the Changes of Muscle Fiber Type

Abstract


Objective: application of enzymes and microscopic image analysis techniques to study changes in mouse gastrocnemius muscle fibers and motor endplates, explore load swimming training on the neuromuscular junction, and muscle fiber. Methods: Male 2-month-old Kunming mice 30 each weight 34 ± 1.87g, national standard rodent diet feeding ad libitum water. Adaptive feeding for one week, were randomly divided into three groups, 10 in each group, the training group training. A. convalesce control group: immersed in the shallow water of the same water temperature, and limit their activities a day, 120 minutes; the B.120-minute group: moderate-intensity swimming training pool volume of 150cm × 32cm × 80cm, 25cm water depth, water temperature of 32 ± 1 ℃, training six days a week, training once a day. The first day of the 10 minutes, the next day for 20 minutes, and thereafter daily increment to the first weekend swimming time reaches 60 minutes reached 120 minutes, the second weekend. Maintain this amount of exercise swimming after eight weeks; C.120 minute negative restructuring: 120 minutes group training conditions and methods of the same, just increase the weight 5% tail (tail) wound with thin wire. Swimming training for eight weeks, with 4% pentobarbital intraperitoneal anesthesia, the speed to take the posterior limbs of the gastrocnemius frozen sections. Enzyme histochemical methods by means of an image analysis system for the measurement and analysis of acetylcholinesterase (AchE), succinate dehydrogenase (SDH), adenosine triphosphatase (ATPase). Results: After 8 weeks of swimming training, the movement of groups of muscle fibers in red muscle fibers of the area percentage compared with the control group has been an increase in red muscle fiber area percentage than in the exercise group and the control group, there is a significant difference (P lt; 0.05); downward trend, the percentage of white muscle fiber area obvious downward trend of the exercise group compared with the control group, there was significant difference (P lt; 0.05). The movement of groups of muscle fibers in red muscle fibers of the diameter compared to the control group have increased significantly, the exercise group and the control group compared to red muscle fiber's diameter, there are very significant differences (P lt; 0.01); decrease in white muscle fiber diameter which sports groups significantly reduce the trend, there is a significant difference compared with the control group. The three sets of motor endplate diameter showed an increasing trend in the control group and the exercise group compared with a significant difference (P gt; 0.05). Image analysis statistics show, compared with the control group, the negative recombinant SDH average integral optical density values ??significantly increased, the difference is very significant with (P lt; 0.01) negative reorganization average integral optical density values ??significantly increase the differences significant with (P lt; 0.05 ). Recombinant the ATPase average integral optical density values ??significantly reduced compared with the control group, the negative, the difference is very significant (P lt; 0.01), no negative restructuring ATPase average integral optical density value decreased, the difference was significant (P lt; 0.05) was statistically significance. Conclusion: swimming training, plus load swimming training can promote changes in skeletal muscle fiber types, and can also expand the diameter of the muscle fibers and motor endplates.
