OBJECTIVE: Polycystic ovary syndrome (PCOS) is the most common women of childbearing age endocrine and reproductive function disorders. Existing studies suggest that PCOS is insulin resistance is characterized by endocrine and metabolic disorders, insulin resistance is not only an important feature of PCOS patients, but also may be the major pathological basis of PCOS, may play a role early in the pathogenesis of PCOS. Calpain-10 gene (CAPN10) is a positional cloning diabetes susceptibility gene, the gene mutation and insulin resistance. The purpose of this study was to explore the the the Shandong Han people calpain-10 gene (CAPN10) single nucleotide polymorphism (SNP-43, SNP-19) with polycystic ovary syndrome (PCOS). Methods: Polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP) detected 111 cases of non-PCOS women and 98 PCOS patients (according to BMI, HOMA-IR divided into obese group and non-obese group, the combined insulin resistance group and combined insulin resistance group) CAPN10 SNP-43 G / A polymorphic SNP-19 Ins / del polymorphic polymorphic genotype distributions were compared between groups, the allele frequency and clinical metabolic parameters. Results: (1) SNP-43 G allele in PCOS and control groups in the distribution frequency were 100% A allele is not found no significant difference between the two groups (P> 0.05); with control compared SNP-19 del / del genotype and del allele frequency increased in the PCOS group (12.3% vs 9.9%, 32.7% vs 30.9%), but the difference was not statistically significant (P> 0.05); (2) SNP-43 GG genotype and G allele frequency obese PCOS group and non-obese PCOS group were 100%, the differences were not significant (P> 0.05), of PCOS merger insulin resistance group combined insulin resistance group were 100%, the differences were not significant (P> 0.05); SNP-19 del / del genotype and del allele frequency obese PCOS group than the non-obese PCOS group (16.0% vs 8.3% , 35.0% vs 30.2%), the differences were not significant (P> 0.05), PCOS and Insulin resistance group than in the non-consolidated insulin resistance group (15.8% vs 7.4%, 34.2% vs 30.5%), the differences were not has significant (P> 0.05); (3) CAPN10SNP-43 G allele frequency in PCOS patients, 100% higher than the foreign study results (76%, 65%), the differences were statistically significant (P <0.05); SNP-19 the del allele frequency in the PCOS group distribution frequency of 32.7%, lower than the distribution in British PCOS patients and African American patients with PCOS (42.5%, 66%), The differences were statistically significant (P <0.05). Conclusion: (1) the calpain-10 gene SNP-43 and SNP-19 with the Shandong Han polycystic ovary syndrome genetic susceptibility no correlation may not be the Shandong Han polycystic ovary syndrome susceptibility gene polymorphic points; (2) SNP-43 G allele and GG genotype and SNP-19 del allele and del / del genotype with polycystic ovary syndrome, obesity and insulin resistance was no correlation; (3) Calcium protease 10 gene SNP-43 allele and the SNP-19 allele distribution racial differences may exist.
