Cotton is one of the most important economic crops in the world, of which Gossypium barbadense has the most superior fiber quality, but the integrated Sea Island Cotton quality in China is low. It offers the possibility to change the status that the transgenic technology of modern plant genetic engineering. Transformation and regeneration of cotton by means of somatic embryogenesis has been widely used in cotton breeding. However, this regeneration system was restricted by genotype. Different developmental stages of different explants in the induction of somatic embryos on callus produce significant differences. Therefore, the selection of specific developmental stage in cotton cells or tissue to induce somatic embryos in the establishment of new regeneration system has become an useful attempt to overcome these problems. Gibberellin (GA) in cotton fiber development has an important function, it can not only promoting fiber initiation but also having effect on cotton fiber diferentiation, elongation,primary wall systhsis cell wall deposition and cotton dwarfism.As a kind of regulator in GA signaling,DELLA negtively regulate GA lever in plant.This green revolution gene have been widely used in crop yield, lodging and other dwarfing in rice, wheat and other crops,which can be used in cotton.Object:To expand genotypes of renewable island cotton in Xinjiang and regenerate the main Island Cotton in Xinjiang through somatic embryo path; achieve genetic transformation of Xinjiang island cotton, and then to improve the integrated quality of sea island cotton.Methods:Ovule of Xinhai 15, a breed of Gossypium barbadense cultured in Xinjiang, were used as explant to figure out a favorable culture system for the regeneration of somatic embryo. pBI121 for the skeleton, two different tissue-specific promoters rbcs2 and rbcs3c in tomato and the gai gene in model plant Arabidopsis thaliana were used to construct two dwarfism vectors. And these two vectors were used to transform the embryogenic callus of XH-15.Result:Make it possible that the regeneration of XH-15 through somatic embryo path; together with favorable treatments,hormones and their concentration in different stages were initially proved. Expansion of renewable island cotton genotypes.Although not realizing the genetic transformation of XH-15, It can help us to improve the quality of Sea Island cotton that the tryinng in genetic transformation by pollen tube pathway.Conclusion:Figured out a favorable culture system for the regeneration of somatic embryo of Xinhai 15; Achieved tissue-specific dwarf expression vectors; not achieving XH-15 genetic transformation due to the suppression of Agrobacterium.
