Abstract


HIV is the human tenacious opponent , AIDS 20 years later , of HIV vaccine research has made important progress ( some vaccine candidates in animal experiments partially blocked HIV infection and delaying AIDS occurred ) since 1981 , but still no successful HIV vaccine research presented unprecedented challenges of today's scientific . HIV has a high degree of variability , the latent infection and immune cells targeting HIV is caused by chronic infection , virus repository established in T cells , macrophages and monocytes , integrated part of the virus in these cells , in order to still proviral form of existence ; virus has a high degree of variability, a strong immune escape ability ; immune cells is the target cells of the virus attack , due to the presence of infection -induced enhanced risk of antibody , and the lack of ideal infection animal model , these are to AIDS vaccine development has brought great difficulties. The target gene Gag-Pol sequence of the conserved region of the HIV-1 epidemic strain , based on codon optimized construct for homologous recombination shuttle plasmid PSC -11 - GagPol detection by homologous recombination and screening to obtain the specific expression of recombinant vaccinia virus HIV-1 Gag-Pol protein rMVA - Psc11 - GagPol after the results of the analysis of humoral and cellular immunity , immunogenicity rMVA-Psc11-GagPol .
