Effects of Self-Assembling Peptide Nanofiber Scaffold on Nucleus Pulposus Cells from Rabbits

Objective: self-assembly of DHL/RADA-16 polypeptide nano fibrous scaffold gel with rabbit nucleus pulposus cells (nucleus pulposus cells, NPCs) joint training , to observe the role of NPCs . Methods : 3-week-old healthy Japanese white rabbits , whichever is the thoracic and lumbar nucleus pulposus , extraction cultured nucleus pulposus cells , nucleus pulposus cells identified with toluidine blue and type II collagen immunohistochemical method . The same amount of cell culture medium DMEM - F12 to trigger the peptide solution the DHL - PA Rada - 16 - PA ( 1:1 ) to self-assemble into DHL/RADA-16 ( experimental group) , the nucleus pulposus cells with self-assembled nano - composite materials training ( experimental group) and nucleus pulposus cells cultured alone (control group) , to observe the self-assembly of nanomaterials on the biological behavior of the nucleus pulposus cells . Results: atomic force microscopy shows DHL/RADA-16 formed nanoscale fibers ; immunohistochemical method for the identification of the nucleus pulposus cells : successfully isolated and cultured nucleus pulposus cells , stained with toluidine blue ( ) , type II collagen antibody ( ) ; DHL / RADA - 16 ( experimental group) nanomaterials with nucleus pulposus cells cocultured , Calcein-AM/PI dead staining of living cells , the number of viable cells more than 95% of CCK - 8 indirect count DHL/RADA-16 can promote myeloid nuclear cell adhesion and proliferation of nucleus pulposus cells without toxic effects . Conclusion : DHL/RADA-16 nanomaterials have good biocompatibility of nucleus pulposus cells can be used as biological material of the nucleus pulposus tissue engineering .
