Abstract

Adenoid cystic carcinoma (adenoid cystic carcinoma, ACC) has the typical characteristics of perineural invasion, the Changcheng jump invasion of peripheral nerve ACC perineural invasion mechanism has very important clinical significance. In preliminary research work by our group, put forward a variety of nerve factor and tumor cell expression of the corresponding receptors in the ACC may histopathological basis ACC perineural invasion, and it may be one of the ACC perineural invasion mechanism. Objective: To observe the research the SD rat sciatic nerve adenoid cystic carcinoma cell proliferation, metastasis and adhesion to understand the role of the sciatic nerve in the culture medium in the process, to study the mechanism of the ACC perineural invasion and perineural invasion control ACC theoretical basis, and at the same time lay the foundation for the clinical diagnosis and treatment for ACC. Methods: SD rat sciatic nerve culture liquid cultured adenoid cystic cancer cells, the Department of the ACCM is MTT proliferation assay sciatic nerve was observed ACC proliferative capacity in vitro culture medium; 2. Cultured SD rat sciatic nerve liquid placed in the the Transwell cell chamber chamber as a chemotactic factor, separate control group chemotactic factors ACC-M cells in the upper chamber, in order to study the impact of the sciatic nerve in vitro migration of ACC-M; application MTT colorimetric the assay SD rat sciatic nerve ACC-M cell culture fluid cultures on the reconstruction of the basement membrane Matrigel adhesion rate, to explore graduate ride: Yang Xiangming Instructor: Professor Sun Moyi bone nerve vitro adhesion ability of ACC-M. Results: 1. Sciatic nerve in the culture medium 48,72 h after OD value of ACC-M cells was significantly higher than the other groups (P lt; 0.01). The description of this group in the ACC-M cell number proliferation significantly. Nerve culture medium as chemoattractant factors ACC-M the number of migrating cells relative to the blank control group, and the co-culture fluid group, there were significant differences (P lt; 0.05), and the number of migrating cells compared with the control group, ACC-M relative to the blank control group increased by 51.5%, while the total medium group only increased by 8.4% compared with the blank control group, did not reflect the significant difference. Reconstituted basement membrane Matrigel remodeling, sciatic nerve adhesion rate of ACC-M cell culture fluid group than blank control group during the same period, and a total medium group (P lt; 0.05), while the control group and a total of no significant difference between the medium group, have certain enhanced cell adhesion after the sciatic nerve were cultured from both negative. Conclusion: SD rat sciatic nerve culture solution can significantly improve the adenoid cystic carcinoma ACC-M cell lines in vitro proliferation ability, able to some extent enhanced ACC-M cells in vitro migration and adhesion. To further infer adenoid cystic carcinoma and perineural invasion, exocrine role of nerve tissue in the tumor cells, thus enhancing their proliferation, metastasis and adhesion ability of this phenomenon may be the histopathological basis of adenoid cystic carcinoma and perineural invasion .
