Abstract


Objective: To investigate the expression of cyclin E expression of P27 KIP1 < / sup> Multi - parameter non phase relationship of the expression with clinicopathological staging of gastric cancer , reveal the law of gastric cancer cell cycle . Methods: This study selected 34 patients with gastric cancer , 10 cases of precancerous lesions and 10 cases of normal gastric mucosa . Mechanical or collagenase and hyaluronidase mixed digestion clinical specimens made ??of a single cell suspension , 80 % the precooling ethanol fixed overnight . 0.5 % Triton X -100 on ice the cells were washed by centrifugation for 5 minutes , FITC-labeled secondary antibodies CyclinE in PE direct standard P27 KIP1 and propidium iodide DNA staining , detected by flow cytometry three - parameter method . At the same time the DI (DNA index ) , PI ( proliferation index ) and SPF ( S - phase percentage ) . Results: 1 . Normal gastric mucosa , precancerous lesions and gastric cancer and adjacent tissues aneuploidy rate of 0,20 % , 76.5% and 17.6% , a significant difference ( P <0.01 ) ; gastric cancer was significantly higher SPF and PI a significant difference . 2. The gastric CyclinE and the expression of P27 KIP1 were 32.08 % ± 15.04% , 5.58 % ± 4.75 % , compared with normal gastric mucosa , precancerous lesions and cancer adjacent tissues , the difference was statistically significant ( P <0.01). Both expression and gender , age , tumor size and Borrmann typing has nothing to do with the depth of invasion , lymph node metastasis , metastasis and TNM stage are closely related ( P <0.05 ) of CyclinE and P27 KIP1 expression was negatively correlated . 34 cases of gastric CyclinE Unscheduled expression was 52.94% , a difference with the control group . Conclusion: Unscheduled overexpression of cyclin E and P27 KIP1 low expression of the G1 / S transition point adjustment out of control , the incidence of gastric cancer , the progress is closely related . Multi-parameter flow cytometry can reveal more comprehensive gastric cancer cell cycle regularity .
