Epilepsy is a common neurological diseases, is caused due to the imbalance of excitatory and inhibitory. Clinical usually antiepileptic drugs (AEDs) in epilepsy produce to suppress seizures, but the general lack of fundamental and effective treatment approaches. At the same time, we know very little about the mechanisms of pathogenesis of epilepsy and clarify the process is very important for the emergence and development of seizures. Important ways and means to create a fast, efficient epilepsy animal models can not only provide mechanisms for epilepsy Mechanism and also the new AEDs screening effective ways. Our previous Camphene chlorothiazide (CTZ) to establish new epilepsy animal models to carry out the preliminary study, we mainly learn through the body field potential technology and behavior observation and pharmacological analysis, key research CTZ and several classical chemical convulsive agent-induced acute epilepsy model in the rat hippocampus, compare the features and characteristics of these models. Based on these models we further experiments, a preliminary study epilepsy pharmacological mechanisms and some antiepileptic effect of the drug, and thus provide a new target for future anti-epileptic drug research. Establish camphene chlorothiazide (CTZ), pentylenetetrazol (PTZ) and kainic acid (KA) model of acute seizures resulting from behavioral studies found significant acute CTZ and KA intracerebroventricular injection epilepsy caused by intracerebroventricular injection of PTZ Whether awake or anesthetized animals lack the ability to cause seizures. Hippocampal electrophysiological model, intracerebroventricular injection of CTZ (5μmol, 5μl) can lead to a gradual increase in the number of time-dependent induced PS peaks (Peaks), with the increase in spontaneous discharge to last synchronization burst-like grand mal The Epilepsy process. Subcutaneous injection of PTZ (383mg/kg) resulted in the CTZ similar epileptiform process. Intracerebroventricular injection of 1μg and 0.5μg KA, resulted in a relatively different seizures. KA rarely induced increase in the number of peak potential, but can quickly form a spontaneous discharge and gradually increased excitability to the short-term, highly synchronized burst-like grand mal, go to more than five minutes of high-frequency continuous seizures (acute status epilepticus (ASE). PTZ model and CTZ model electrophysiological recording process found that there is a high degree of consistent synchronization phenomenon between different brain regions of the ipsilateral hippocampus during seizures. pharmacology studies have shown that benzodiazepines class of tranquilizers alprazolam (0.4mg/kg, ip) significantly inhibited the CTZ and PTZ-induced epilepsy role, but the role of the KA is not obvious. noncompetitive AMPA receptor antagonist GYKI 53784 (5mg/kg iv) only inhibit CTZ caused by burst grand mal and not inhibit PTZ-induced burst PTZ model-based study found, methanol can suppress the intensity of seizures, the antiepileptic effect of this study suggest that chemically induced epilepsy model has a certain mechanism electrophysiological representation of the same sex and different, while our laboratory to establish the CTZ-induced epilepsy model has special features for the pathogenesis of epilepsy and antiepileptic drug screening provides a good tool.
