Purpose: adipokines and inflammation of the recent attention to its role in arthritis is a whole new topic. Our previous study also found that the adipokines adiponectin in the synovial fluid of rheumatoid arthritis (RA) (AD) was significantly higher than the level of bone arthritis (OA). Our partners found another a fat factor heparin-binding epidermal growth factor (HB-EGF) gene significantly higher expression in ankylosing spondylitis chip analysis. The purpose of this experiment is to study the role of AD and HB-EGF in RA chronic persistent inflammation and bone destruction in, explore its significance may exist, and to lay the foundation for further experiments. Methods: (1) were measured by ELISA 62 RA patients and 22 age, gender, body mass index (BMI)-matched healthy volunteers serum AD and HB-EGF protein level, and at the same time line Real Time-PCR detection of peripheral blood mononuclear mononuclear cells (PBMC) AD and HB-EGF mRNA content. (2) ELISA determination of RA and OA synovial fluid of AD and IL-1β, IL-6, and the content of pro-inflammatory cytokines such as TNF-α, analysis of AD and its relevance. Results: (1) RA severe active group in the serum of patients with AD and HB-EGF levels (15.8 ± 2.7 ug / ml, 745.9 ± 217.0pg/ml) were significantly higher than moderate (13.8 ± 2.6 ug / ml, 715.8 ± 189.3 pg / ml), low activity group (14.9 ± 1.4 ug / ml, 575.4 ± 75.1 pg / ml), remission group (13.4 ± 2.1 ug / ml, 561.5 ± 80.5 pg / ml) and healthy control group (13.5 ± 2.1 ug / ml, 492.2 ± 59.9 pg / ml) (P lt; 0.05), while correlation analysis found that AD HB-EGF DAS28, erythrocyte sedimentation rate (ESR), rheumatoid factor (RF) or C-reactive protein ( CRP) were positively correlated (r = 0.3-0.5, P lt; 0.01). But each group PBMC was not detected to the AD mRNA, although the PBMCs in the HB-EGF mRNA can be detected, but no significant differences among the groups. (2) RA synovial fluid of AD, IL-1, IL-6, TNF-α content compared with OA high, the AD and IL-6 positively correlated (r = 0.27, p lt; 0.05). (3) AD and IL-6 co-stimulation, MH7A RANKL mRNA expression was significantly increased. (3) successfully wrapped the rAAV2-prepared with Adiponectin. the rAAV2-adiponectin successful transfection of THP-1 cells, but not detected inflammatory cytokines (IL-1, IL-6, TNF-α) change. Conclusion: AD and HB-EGF may be involved in chronic inflammatory processes of rheumatoid arthritis, further study of AD and HB-EGF and sources involved in the pathophysiological process may further clarify the pathogenesis of RA, and targeted treatment of RA provide new ideas.
