The purpose of the hepatitis B virus (HBV) infection is the most important risk factors for human liver cirrhosis and hepatocellular carcinoma. In this study, a high-speed large number of injections containing HBV genome eukaryotic expression plasmid production HBV infection model via the tail vein, and injection of HBV siRNA, to detect interference effect, to lay the foundation for further study of its interference with immune effector mechanisms. Method 1 through the tail vein high-speed, large number of injections containing HBV genome eukaryotic expression plasmid production HBV infection model, the fourth day of treatment, respectively dosage effect relationship, the relationship between aging and the three imaging system observed plasmid pGenesil-hk GFP expression , verify that the model is successful. 2. Using time-resolved fluorescence assay siRNA interference mouse serum before and after the secretion of hepatitis B surface antigen (HBsAg) and e antigen (HBeAg), total RNA from liver cells using real-time quantitative PCR, immunohistochemical detection of liver tissue HBcAg. Results of a model of HBV infection in mice at high speed through the tail vein injection of HBV expression plasmid after four days, the quantitative detection of the HBsAg and HbeAg results show that pBlue-HBV gradually increased the amount of HBeAg rise; liver cells flow cytometry 2-9 days have varying degrees of expression, 3-5 days is the expression of the peak period. The results of in vivo imaging of mouse liver expression of green fluorescent normal mice, no fluorescence signal. Thus, we can confirm that the high-pressure water power established by injecting HBV infection in a mouse model, expression of HBV, serum liver? Detected HBsAg and HBeAg, and peak expression 3-5 days for about 10 days. 2.HBV siRNA interference effect Realtime PCR results show that siRNA group significantly inhibit the expression of HBsAg and HBeAg. Constructed the siRNA in vivo effectively inhibited at the mRNA level expression of HBV; Time-resolved fluorescence assay results suggest pGenesil-1 and pGenesil-2 group HBsAg and HBeAg expression decreased, indicating that siRNA can effectively inhibit the expression of HBV-associated antigen, further immunohistochemical detection results show that the undisturbed group showed more positive cells in pGenesil-1 and group 2 is almost no positive cells, siRNA can effectively inhibit the expression of HBcAg. Conclusions by high pressure hydrodynamic injection successfully established HBV infection in a mouse model, hepatic expression of HBV, HBsAg and HBeAg detected in the serum. The same method of injection HBV siRNA can effectively inhibit HBV mRNA and antigen expression, and the expression of HBcAg in liver cells, and laid the foundation for further in vivo targeting of interference with immune effector mechanisms in vivo.
