Abstract


Ulcerative colitis (ulcerative colitis, UC) is a cause is not yet clear of the rectum and colon chronic non-specific inflammatory disease. UC pathogenesis is not clear, generally considered related to factors such as environment, heredity, infection, immunization, smoking and mental. Now that the immune abnormalities, especially knot immune disorder of the intestinal mucosa partial occupies an important role in the pathogenesis of UC, and promote an increase in inflammatory cytokines and anti-inflammatory cytokines relative reduction is caused by colonic mucosa abnormal immune response and chronic inflammation reasons. Transforming growth factor-β (transforming growth factor-β, TGF-β) is a kind of cell growth, differentiation and a variety of physiological and pathological processes, cytokines play an important regulatory role, and is a member of the TGF-β superfamily. TGF-β as an important anti-inflammatory cytokines by down excessive immune response and inhibit inflammation and development. TGF-β in vertebrates has been found that the five kinds of subtypes (TGF-β1 to 5), but in mammals mainly presence of TGF-β1-3. Objective: method of the experiment using immunohistochemical detection UC with irritable bowel syndrome (irritable bowel syndrome, IBS) patients expression knot of TGF-β in the intestinal mucosa and its subtypes, to investigate the TGF-β and its subtypes with the the UC relationship and explain its role in the pathogenesis of this disease. Methods: 50 patients with UC and 30 cases of IBS colonoscopy biopsy wax block specimens for research pedigree. By immunohistochemical ELPS (enhance labeled polymer system) method detects two TGF-β isoforms expression in colonic mucosa lesions; observe different disease stage, different severity, different clinical types, extent of disease UC TGF -β isoforms; analysis of various subtypes of TGF-β and the TGF-β expression in colonic mucosa in UC. Results: 1.TGF-β expression in the UC group significantly stronger than the IBS group (P lt; 0.01) and the active phase was significantly higher than in remission (P lt; 0.01). In active UC, TGF-β expression in the different clinical severity between the two groups have a significant difference (P lt; 0.05); different clinical types, different lesions between the two groups were not statistically different (P gt; 0.05) . 2.TGF-β1 expression in the UC group was significantly stronger than the IBS group (P lt; 0.01), and the activity was significantly higher than in remission (P lt; 0.01). In active UC, the expression of TGF-β1 in the different clinical severity between groups statistically significant difference (P lt; 0.05); different clinical types, different lesions between the two groups were not statistically different (P gt; 0.05) and was positively correlated with the expression of TGF-β (rs = 0.749, P lt; 0.05). 3.TGF-β2 expression in the UC group was significantly stronger than the IBS group (P lt; 0.01) activity was significantly higher than in remission (P lt; 0.01). In active UC, the expression of TGF-β2 in the different clinical severity between groups statistically significant difference (P lt; 0.05); different clinical types, different lesions between the two groups were not statistically different (P gt; 0.05) and was positively correlated with the expression of TGF-β (rs = 0.690, P lt; 0.05). 4.TGF-β3 in the UC group and the IBS group showed no significant difference (P gt; 0.05), and the expression of activity and remission there was no significant difference (P gt; 0.05). In active UC, the expression of TGF-β3 in different severity of different clinical types, extent of disease between the two groups were not statistically different (P gt; 0.05), and the expression of TGF-β was no correlation (rs = 0.068 , P gt; 0.05). Conclusion: colonic mucosa in UC With aggravation positive expression of TGF-β increased significantly, associated with the pathogenesis of UC, so monitoring of UC junction TGF-β in the intestinal mucosa can understand illness staging of the disease and the serious level. In UC adjustment mechanism involved in the inflammation through the secretion of TGF-β, TGF-β1, TGF-β2 play an important role. 3 in UC adjustment mechanism involved in the inflammation through the secretion of TGF-β, TGF-β3 may not play a significant role.
