Objective:The atherosclerosis was induced in rabbits by fed a high cholesterol diet and the effect of Yi Xin Shu on experimental atherosclerosis and endothelial function studies in rabbits was evaluated.Methods:The 24 New Zealand white rabbits were randomly divided into three groups of eight, the first group to a normal diet; second group were fed a high cholesterol diet; third group with Yi Xin Shu (1g/d/Kg), at the same time the rabbits were fed a high cholesterol diet for 4 months.the atherosclerosis, VACM-1 and endothelial function was evaluated by HE staining, oil red O staining,immunohistochemical staining and detection of endothelium-dependent vasodilation.Results:1. Comparison of three groups of lipids:The degree of atherosclerosis was significantly lower in Yi Xin Shu group than that in high-cholesterol group.2. Three degree of atherosclerosis in comparison:The intima thickness and intima-media thickness ratio (I/M thickness) of Yi Xin Shu Group were significantly reduced than the high-cholesterol group(P<0.05).3. Three groups of atherosclerotic plaque lipid changes:Yi Xin Shu group lipid content was less than high-cholesterol group (P<0.05);4.Three groups of atherosclerotic plaque VCAM-1 protein expression:VCAM-1 was mainly expressed in endothelial cells, high-cholesterol group compared with Yi Xin Shu group was significantly increased (P<0.05), while the PBS negative control group, no expression.5. Three groups of endothelium-dependent vasodilatation function in comparison:Yi Xin Shu is higher than the high-cholesterol group (P<0.05), but lower than the normal(P<0.05).Conclusions:The treatment with Yi Xin Shu significantly reduced the degree of atherosclerosis, decreased expression of VCAM-1 protein, and improved the function of endothelium-dependent vasodilation.
