Abstract


Abstract:Objective To investigate effects of Dendrobium nobile Lindl. alkaloids (DNLA) in diabetic rats induced by high-fat diet and streptozotocin. Methods Male Spragye-Dawley rats were randomly divided into 5 groups:control, model, metformin (100 mg/kg), DNLA (20 mg/kg,40 mg/kg). Male rats were fed with a diet enriched with triglyceride, sucrose and cholesterol to induce insulin resistance for 6 weeks. Streptozotocin 35 mg/kg was then injected intraperitoneally to induce hyperglycemia, a model similar to the metabolic abnormalities of human type 2 diabetes mellitus. After intragastric administration of metformin or DNLA for 4 weeks, the fasting insulin (FINS), glucacon-like peptide 1 (GLP-1),caspase3, tumor necrosis factor-a (TNF-a) were determined. The expression of insulin (INS) protein was examined by immunohistochemistry (IHC). Results The level of FBG was increased in model group, and the levels of FINS, GLP-1 were decreased, but caspase3 and TNF-a were significantly increased in model rats. A decrease in number and size of pancreatic island was observed. DNLA (20 mg/kg,40 mg/kg) enhanced the levels of FINS and GLP-1,meanwhile significantly attenuated the elevations of caspase3 and TNF-a, furthermore pancreatic islet cells damage was mild as compared with model group. The expression of INS protein in pancreatic islet was significantly decreased in model group. DNLA at high and low doses significantly increased the INS above mentioned. Compared with model group the down trend of FBG in Metformin group and DNLA groups was not obvious. Conclusion DNLA can significantly increase the levels of INS in diabetic rats and protect pancreatic island beta cells. It can reduce the expressions of caspase3 and TNF-a in diabetic rats. Methods:KKAy mice(FBG>7.0mmol/L) were randomly seperated into 4 groups (n=5):control, model, metformin (1.5mg/10g), DNLA (20mg/kg, 40mg/kg), and control group included 5 C57BL/6J mice, female and male groups were even and in common. After intragastric administration of metformin or DNLA for 2 weeks, the fasting blood glucose (FBG), triglyeride (TG), cholesterol (CHOL), AST/ALT, the ratio of high density lipoprotein cholesterin and low density lipoprotein cholesterin (HDLC/LDLC) were detected respectively. Results:In male groups, compared to the control group, the levels of FBG, TG, CHOL were significantly increased in model mice; Compared to the model group, Metformin, DNLA attenuated the elevations of FBG, TG,.Metformin and DNLA (40mg/kg) had the trend to decrease CHOL. The ratio of AST/ALT was not increased in other four groups compare to the control group. The ratio of HDLC/LDLC was significantly decreased in model control, and increased obviously in Metformin group, DNLA groups were increased slightly. In female groups, compared to the control group, the level of FBG was obviously increased in model group; Metformin and DNLA could decrease the level of FBG in KKay female mice mildly, but the effect was not obvious compared to which in male mice. The rest of biochemical indicators were similar in female and male groups. Conclusion:DNLA could effectively reduce the level of fast blood glucose and regulate glucolipid metabolisms in diabetic KKay male mice.
