Abstract
|
Colorectal cancer is one of the most common human malignancies, its incidence in developed countries accounted for 2-3 of all malignancies in our first 4-5. In recent years the incidence of colorectal cancer, invasion, transfer mechanism, has become a hot research scholars. Nuclear factor kappa B (NF-κB, nuclear factor of kappa B) is a nuclear transcription factor found in recent years, can regulate the expression of many genes at the transcriptional level in the body's immune response, inflammatory response and cell growth and development other aspects play an important role, and its relationship with the tumor by more and more attention from scholars. Although studies have shown that the expression and activity of NF-κB in tumor cell lines and tumor tissues were increased, but is still controversial in the relations of NF-κB and tumor differentiation, lymph node metastasis, clinical stage, and domestic NF-κB and relationship between clinicopathological parameters of colorectal reports are still rare. This topic using immunohistochemistry En Vision two-step detection of colorectal carcinoma and adjacent tissues in the NF-kappa B p65 subunit protein expression, and to explore the relationship between NF-κB and colorectal cancer clinical pathology parameters to a period of early diagnosis and prognosis of colorectal cancer to develop a reasonable comprehensive treatment program to provide new ideas and methods. Materials and methods: randomly selected First Affiliated Hospital, Zhejiang University School of Medicine Anorectal March 2001 - 2001 11 menstrual pathologically confirmed colorectal carcinoma 65 cases, 42 cases of cancer tissue. The immune histochemical En Vision Zhejiang University, Master's thesis two-step testing organization NF-B p65 protein expression. Judge the yellow finely granular cytoplasm precipitation nonspecific staining intensity above background staining positive expression, and Reln! The immune response nele and Stegner integral (workers RS) scoring method for semi-quantitative analysis. Statistical analysis using x 'test. Results and discussion: NF-KB p65 protein expression in colorectal cancer (P lt; 0 .01) was significantly higher than the adjacent tissues. NF-KB p65 protein expression with tumor differentiation, poorly differentiated colorectal cancer group was lower than in well-differentiated colorectal cancer group (P lt; 0 .05). NF-B p65 protein expression with age, gender, tumor size, lymph node metastasis, the tumor DukeS staging. Nuclear factor KB was first discovered in 1986 by Sen and the Bal seven imore, many studies have shown that NF-KB in cell survival, the salary attached cells, cell differentiation, cell growth and plays an important role in the inflammatory response, has been confirmed clever 0 substances can activate NF-KB activation of NF-KB can activate more than 150 kinds of genes, and found many with cell transformation, proliferation, tumor invasion, angiogenesis related to tumor-related genes governed by NF-KB regulation . This experiment NF KB p65 protein expression in colorectal cancer was significantly higher domestic and international study found that colon cancer cell line abnormally high expression of NF-KB, that NF-KB in the incidence of colorectal cancer, growth plays an important role. This experiment NF KB p65 proteins in poorly differentiated colorectal tissue expression lower than that of the well-differentiated organization, access to research at home and abroad, most of that was no significant difference in well-differentiated and poorly differentiated expression of individual that poorly differentiated expression of higher for medium well differentiated. Consider each experiment there are experimental methods selected specimens, the impact of the differences on the the selected reagent sensitivity and specificity, and statistical methods, to be confirmed by further study. NF in the experimental KB p65 protein expression in colorectal cancer lymph node metastasis, consider NF-KB is the extensive role of nuclear transcription factor, NF-KB is quite high intensity of expression had emerged early in tumorigenesis, immunohistochemical methods may unable to further distinguish between lymph node metastasis in colorectal cancer tissue differences in the expression, try other quantitative testing methods. Conclusions: 1) the NF a KBp65 protein in colorectal cancer increased expression. Zhejiang University, Master Thesis 2) the degree of differentiation of the the NF a KBp65 protein expression and colorectal cancer was positively related. 3) NF KB p65 protein expression in colorectal cancer patients age, gender, tumor stage, tumor size, lymph node metastasis. 4) NF KB p65 protein detection can be used for the detection of colorectal cancer, as in colorectal cancer diagnosis, therapy an objective indicators.
