Background and Purpose:Colorectal cancer is common in the human digestive tract malignant tumor and themorbidity and mortality increased year by year, which constitutes a serious threat to humanhealth and life. Colorectal cancer early diagnosis rate is low, recurrence and metastasis is amajor cause of death. Molecular mechanism of gene in most of the current domestic andinternational medical researchers focus on the study of colorectal cancer, in order tofacilitate early diagnosis and targeted therapy of colorectal cancer and reduce mortality.Fragile histidine triad gene is a candidate tumor suppressor gene, mainly realizes its tumorsuppressor role through participation in the regulation of cell cycle and apoptosis induction.CyclinDl is the G1phase of the cell cycle, the expression once out of control, will causethe cell cycle disorder caused by tumor, is therefore regarded as the cancer gene. Thepurpose of this study was to explore the expression of FHIT and CyclinD1in normalcolorectal mucosa, atypical hyperplasia, and colorectal cancer, to explore the relationshipbetween the clinical and pathological characteristics of these two kinds of protein withcolorectal cancer and their correlation expression in the large intestine. And then analyzetheir role in colorectal carcinogenesis, development, invasion and metastasis. The resultmay provide theoretical and experimental basis for clinical diagnosis and treatmentprognosis better.Methods:FHIT and CyclinD1expression are assessed immunohistochemistry S-P in90cases ofcolorectal cancer,36cases of atypical hyperplasia,30cases of normal colorectal mucosa.Then do the statistical analysis of the relationship between the positive expression incolorectal cancer and their clinicopathological features.Specimens of colorectal cancer aredivided into groups based on parameters such as sex, age, tumor location, degree ofdifferentiation, depth of invasion, TNM stage, lymph node metastasis and so on. Thencalculate the positive rate and compare them.The data was statistical analyzed by softwareof SPSS13.0, probability below0.05is specified to be statistics significance. Results:(1) The positive expression of FHIT in colorectal cancer group was51.1%(46/90),significantly lower than the positive expression rate of atypical hyperplasia group72.2%(26/36) and the positive expression rate of the normal control group93.3%(28/30), therewas statistically significant difference between the three (P<0.01).The positive expressionof CyclinD1in colorectal cancer group was60.0%(54/90), significantly lower than thepositive expression rate of atypical hyperplasia group38.9%(14/36) and the positiveexpression rate of the normal control group0%(0/30), there was statistically significantdifference between the three (P<0.01).(2) The decrease or lack of FHIT protein expression in colorectal cancer wassignificantly correlated with its degree of differentiation, TNM stage, lymph nodemetastasis(P<0.05), but there was no correlated with sex, age, tumor location, depth ofinvasion(P>0.05). The enhanced expression of CyclinD1protein in colorectal cancer wassignificantly correlated with its degree of differentiation, lymph node metastasis, TNMstage,(P<0.05), but there was no correlated with sex, age, tumor location, depth ofinvasion(P>0.05).(3) there was the negative correlation between FHIT expression and the expression ofCyclinD1in colorectal cancer (r=-0.539, p=0.000).Conclusion:1. The decrease or lack of FHIT expression is closely related with colorectal cancer,and may be involved in the regulation of apoptosis in colorectal cancer cells, so it can beconsidered as a potential diagnostic marker of colorectal cancer.2. CyclinD1expression was significantly increased in colorectal cancer, suggestingthat it may be associated with colorectal cancer growth, metastasis, deterioration and aclose relationship between the prognosis. CyclinD1is a kind of cell protein, whoseexpression out of control will result in disturbance of cell cycle, directly influence thedevelopment and progression of colorectal cancer.3. FHIT expression was negatively correlated with CyclinDl protein in colorectalcarcinoma, suggesting that the two effects in the development of colorectal cancer survivalmay be the opposite effect. Expression levels of FHIT and CyclinD1combined detectionof pathology for colorectal cancer early diagnosis, evaluation of the biological behaviorand prognosis of colorectal cancer will provid necessary basis of immunology.
