1. Establishment of animal model with Tympanosclerosis in guinea pigs during chronical otitis media.To investigate the courses and relevant pathological changes of Staphylococcosis otitis media in guinea pigs, staphylococcosis were inoculated into the middle ear of guinea pigs, and the guinea pigs were sacrificed periodically, the changes of their middle ears were reviewed by microscope,in order to understand the hearing threshold and the change of tympanosclerosis in middle ear mucosa. All inoculated ears from day 7 contained mucosal adhesions of various sizes, shapes, and locations. The site of predilection for the development of tympanosclerosis was the hypotympanum, followed by the anterior epitympanum, the attic, the drum, the interossicular spaces, and the tubal orifice.The pathological changes are similar to those of human being.lt is a good animal model to study tympanosclerosis during chronical otitis media.2. Expression of Bone Morphogenetic Protein in middle ear mucosa of the guinea pig.Immunohistochemical ABC method is undertaken to explore the expression of Bone Morphogentic Protein (BMP) in the middle ear mucosa of guinea pigs.In the normal middle ear mucosa, In the normal middle ear mucosa, the BMP positive cells distributed on the basement membranes and neovasculations. The results indicated the BMP possibly plays an important role in the middle ear mucosa, and it may play a certain role in the development of middle ear.3. Expression of Bone Morphogenetic Protein and Fibronectin in middle ear mucosa of the guinea pig with chronic suppurative otitis mediaTo explore the expression of bone Morphogenetic Protein (BMP) and Fibronectin (FN) in middle ear mucosa of the guinea pig with chronic suppurative otitis media and to supply the exparimental data for the role of BMP and FN in the Chronic otitis media and tympanosclerosis. 25 healthy guinea pigs were employed in the experiment. After general anaesthesia, by infecting 1 106 Staphylococcosis aureus into the middle ear cavities of right ears with the left ears serving as controls, the supprative otitis media model was established.Then 45 days later, immunohistochemical ABC method was undertaken to explore the BMP and the FN expression in middle ear mucosa of the guinea pigs using BMP monoclonal antiboby and FN monoclonal antiboby. hi the normal middle ear mucosa, the BMP positive cells and the FN positive cells distributed on the basement membranes and neovasculations.The presence of inflammation in middle ear induced congestion, oedema, haemorrhage, and the positive cells of BMP and FN increased. The results indicated the BMP and the FN possibly play important roles in the mechanisms of otitis media development, and it may play a certain role in the occurrence of tympanosclerosis.4. Expression of Bone Morphogenetic Protein and Fibronectin in middle ear mucosa of the patients with chronic suppurative otitis mediaTo explore the expression of bone Morphogenetic Protein (BMP) inmiddle ear mucosa of the patients with chronic suppurative otitis media and to supply the exparimental data for the role of BMP and FN in the Chronic otitis media and tympanosclerosis. Immunohistochemical ABC method was undertaken to explore the BMP and the FN expression in middle ear mucosa of the patients with chronic suppurative otitis media using BMP monoclonal antiboby and FN monoclonal antiboby, with 9 healthy middle ear mucosa serving as controls. In the normal middle ear mucosa, the BMP positive cells and the FN positive cells distributed on the basement membranes and neovasculations. The presence of inflammation in middle ear induced congestion, oedema, haemorrhage. The results indicated the BMP and FN possibly both play important roles in the mechanisms of otitis media development, and it may play a certain role in the occurrence of tympanosclerosis.
