Objective: To study the cartilage repairing effect with allogenic chondrocyte/ PDLLA graft directly, the way may provid experimental and theoretic evidence for allogenic chondrocyte/ PDLLA graft to repair the defects of ear, nose, throat and tracheae directly.methods:1.Study on repairing the cartilage defects using TGF-β1 allogenic chondrocytes/ PDLLA graft: A total of 18 rabbits were divided into the TGF-β1. and chondrocytes/ PDLLA graft group,the chondrocytes/ PDLLA graft group, and the blank control group .Samples were taken out at 4,12 and 18 weeks after implantation .The histological characteristics were investigated.2. The Experimental study on neocartilage formation using allogenic chondrocytes in vivo directness: Obtain the allogenic rabbit ear chondrocytes within one month years old.After detaching and digesting chondrocytes in vitro,they were seeded onto the PDLLA polymers graft, and then implanting subcutaneously of the rabbit after 6 hours . After 6,12 and 18 weeks , thehistological characteristics were investigated.3.Study on repairing the cartilage defects using the allogenic chondrocytes/ PDLLA graft directly: Obtain the allogenic dog thyroid chondrocytes .After 2 hours of detaching and digesting chondrocytes in vitro,they were used to repair the dog thyroid defect , the PDLLA without cells were served as control. Samples were taken out at 4 and 12 weeks after implantationthe ,the histological characteristics were investigated.Results:1. Repairing the cartilage defects using TGF-β1. allogenic chondrocytes/PDLLA graft: After 18 weeks of the transplantation , the repaired effectsof TGF-β1. and allogenic chondrocytes/ PDLLA graft group was betterthan the allogenic chondrocytes/ PDLLA graft group not only on generalsample but also histology ,the blank control group were repaired by fibroustissue.2. Formation: After 6 weeks of implanting, it was founded that the matrixsecreted and there were new and no-mature chondrocytes in the chondrocyte/PDLLA polymer. When 12 weeks of implanting, chondrocytes were close tomature and form cave.After 18 weeks, neocartilage was close to normalcartilage.3. Repairing the cartilage defects using the allogenic chondrocytes/ PDLLAgraft directly: After 4 weeks of experimental group, the defect area wererepaired by kinsmanship cartilaginous tissue; After 12 weeks of implanting,regenerate cartilage tissue filled of the defect area. But the control group wasrepaired by fibrous cartilage or fibrous tissue.Conclusion:1. The way through graft of allogenic chondrocyte/ PDLLA polymer with higherporosity is a promisimg way for repairing the rabbit ear cartilige defects.The allogenicchondrocytes/ PDLLA graft could repair the rabbit ear cartilige defects ,andthe way using TGF-β1 could improve the quality of the rabbit ear cartiligedefects.2. It is feasible to form neocartilage with the way of the allogenic rabbitchondrocytes in vivo directly.3. The allogenic dog chondrocytes/ PDLLA graft may repair the dog thyroiddefects directly,the way may offer theory foundation for clinic emergencytreatment of the head and face cartilage defectly .Significance:This study can provide the theoretic and experimental basis for the allogenic chondrocytes/ PDLLA graft using in clinic emergency treatment of the head and face cartilage defects directly.
