Abstract


Gosling plague (Gosling Plague, GPa) by Parvoviridae Goose Parvovirus Parvovirus genus (Goose Parvovirus, GPV) caused a deadly infectious diseases. The virus is mainly to make the 1-month-old goslings and Muscovy Duck an acute, highly contagious, septic diseases, infectious and high mortality. Closely related to the hazards and age, the 10-day-old goslings incidence and mortality rates of up to 100%, up to 70% mortality in susceptible groups, onset after increases gradually reduce with age. But in recent years has been reported that the incidence of the disease-day-old increasing trend for more than 30 days of age accounted for 14.3%, which may be enhanced with goose parvovirus virulence and genetic variation. One found for the first time since 1956, China scholars Fangding and isolate the virus so far, at home and abroad have been isolated reports of the virus, the disease already was a world-wide epidemic, a great threat to the goose industry. The purpose of this test is to research Goose Parvovirus antibody ELISA method for detection of, and initially used in field trials. Gosling plague virus generally does not cause the large goose incidence, big goose can render latent infection, continued infected, detoxification, resulting in levels of the virus to spread. Gosling plague serum antibody detection will not only be able to monitor the goose health status, but also be able to detect the vaccine's antibody levels, especially the the the goose detection of latent infection infected, to reduce a big goose vivo strain level or vertical transmission a certain sense. Far gosling plague antibody detection and abroad reported the agar diffusion test and neutralization test, but many scholars reported that the sensitivity of the agar diffusion test is not high, and high titer antiserum and concentrated allantoic fluid, in complicated operation and the test takes longer, costs big drawback is not easy to promote practical applications, in order to make up for the lack of the two test methods for detection gosling plague serum antibody, and the establishment of an indirect ELISA method to detect GPV antibodies for surveillance and epidemiological investigations of vaccine antibody levels to provide a certain basis. Test duck embryo proliferation goose parvovirus chloroform treatment, differential centrifugation, PEG precipitation, Sephrose-4B column chromatography treatment, purification of virus antigen. Attenuated live vaccine healthy adult geese preparation hyperimmune serum with the antigen and buy gosling plague as the reference positive serum ELISA test, the detection ELISA OD values ??greater than 1.0. Reference negative serum collected from different days different species from a remote non-infected areas of rural gosling about 20 days of age, 50 non-immune serum the detection ELISA OD value is less than 0.3.
