Abstract


[Background] the past 20 years, China's female breast cancer incidence is rising rapidly and the patient's age was young trend has become one of the most important malignant threat to women's health and life. From a new perspective affects the risk factors for breast cancer, has become a cause of breast cancer the field of exploration and prevention urgent need to address the issue. Studies have shown that estrogen and progesterone and its related factors closely related to breast cancer, estrogen receptor (ER, estrogen receptor), progesterone receptor (PR, progestin receptor) gene polymorphism may be estrogen and progesterone cause breast an important factor in cancer carcinogenic chain. [Objective] The purpose of this study is to explore the estrogen, progesterone receptor gene polymorphism (ER codon325, PR 331G / A) whether to raise the risk of breast cancer of the female population in China; between two loci and in-depth study and their gene - gene and gene - Setting the combined effects of environmental factors in the etiology of breast cancer research and screening of high-risk populations cumulative item. [Methods] In this study, cases for January 1, 2005 to 1 July 2006 in General Surgery, Changzhou First People's Hospital Breast Branch and the First People's Hospital of Suzhou new pathologically confirmed breast cancer cases, aged 25 to 70 years old. ER codon325 polymorphic loci CC, CG and GG genotype frequency distribution in the general population of women, respectively, to 25.23%, 53.27%, 21.50%, PR 331G / A multi-polymorphic loci GA and GG genotypes in women The population distribution of frequencies were 92.99%, 7.01%, respectively. A single gene locus and breast cancer: study found no significant association between the ER codon325 polymorphic loci associated with breast cancer, compared with the CC, CG and GG genotypes adjusted OR was 0.99 (95% CI: 0.61- 1.61) and 1.13 (95% CI :0.62-2 .04). The study found that PR 331G / A polymorphism showed a trend of increased risk of breast cancer, compared with GA, the GG genotype OR = 1.51 (95% CI :0.74-3 .07), but not significant. On the joint genetic risk estimates, the study found that ER codon325, PR 331G / A joint polymorphism with increased risk of breast cancer trends, but not significant. With a history of spontaneous abortion and women carrying the CC genotype compared PR 331G / A polymorphism and found no history of spontaneous abortion and carry CG and GG genotype women OR 3.57 (95% CI :1.21-10 .52); environmental risk factors for breast cancer risk dangerous combination. [Conclusion] 1. Early age at menarche and history of taking oral contraceptives to women with breast cancer risk factors; a tea years long protective factors. 2. There was no significant correlation between ER codon325 polymorphism and breast cancer; 3. Weaker correlation may exist between the PR 331G / A and breast cancer; 4. ER codon325 PR 331G / A between there may be increased risk of breast cancer combined effects; 5. Between the the ER codon325 polymorphism and spontaneous abortion increase the risk of breast cancer combined effects close to the level of significance.
