Abstract

The the florfenicol nanoemulsion The purpose of this study is to develop a high encapsulation efficiency, good stability, safe, efficient, and study its effect on the treatment of mastitis in dairy cows. 1 Florfenicol nanoemulsion this experiment was prepared by poly polyoxyethylene hydrogenated castor oil (RH-40), Tween-80, the SECRETARY classes -80 surfactant; to n-butanol, propylene glycol, glycerol, water, ethanol and the like as a cosurfactant; liquid paraffin, soya bean oil, isopropyl myristate, propyl (IPM), ethyl acetate, and castor oil, glacial acetic acid, such as the oil phase to prepare a blank nanoemulsion. Investigated different surfactant, cosurfactant, by drawing a pseudo-ternary phase diagram, the oil phase, water phase, Km value nanoemulsion forming region, and combined with Origin7.5 software analysis, screening the best carrier drug formulations. The results showed that the nanoemulsion area of ??drug-loaded system, can be used as prescription Florfenicol nanoemulsion Tween-80 / glycerol / ethyl acetate / distilled water. 2. Physical and chemical properties of florfenicol nanoemulsion investigated by transmission electron microscopy, laser particle size distribution analyzer to its physical and chemical nature of the visit. The results show that, Florfenicol nanoemulsion is pale yellow, homogeneous, transparent, good liquidity, visible light blue opalescent liquid, the average particle size (30 ± 5) nm, narrow particle size distribution range and relatively uniform, in line with the requirements of the nanoemulsion. 3. Investigation of florfenicol nanoemulsion stability was measured by UV spectrophotometer the Florfenicol nano milk Florfenicol test content, test by centrifugation, thermal stability testing, accelerated testing and illumination test visits its stability. The results show that, Florfenicol has an absorption maximum at 254 nm, a good linear relationship in the range of 1μg/mL ~ 50μg/mL, the average recovery was 98.3%, RSD was 0.41%, the average encapsulation efficiency was 88.08 ± 0.22 %. The method can be used for the encapsulation efficiency and Determination of florfenicol nanoemulsion; centrifuge test and thermal stability test, accelerated test, appearance and content were not changed significantly. 4 the Florfenicol nanoemulsion security evaluation of this test by the acute toxicity experiments on mice, rabbits skin stimulation test and eye irritation test to Florfenicol nanoemulsion security visits. The results showed that the the Florfenicol nanoemulsion LD50 for mice to 1949.8 mg / kg, there was no significant stimulation of rabbit skin and eye. 5. The Florfenicol nanoemulsion vitro antibacterial test were selected to cause mastitis in dairy cows three common pathogens in vitro antibacterial test tube dilution method twice florfenicol nanoemulsion. The results show that, of nanoemulsion FLORFENICOL on pathogenic E. coli, Streptococcus MIC and MBC is florfenicol solution of 1/2 times; Staphylococcus aureus MIC and MBC florfenicol solution 1/4 times. Mastitis in dairy cows. Florfenicol nanoemulsion treatment trials in this study by the determination of the emulsion before and after the treatment of mastitis somatic cell count (SCC), the results show the of florfenicol nanoemulsion treatment of mastitis in dairy cows better than florfenicol injection. Of milk chamber perfusion a 50 mL FLORFENICOL nanoemulsion, once a day, continuous infusion 8 d good therapeutic effect of mastitis in dairy cows.
