Abstract


TC21/R-Ras2, one member of the four ras supergene families, is associated with growth, migration and transformation of tumor cells. Previous studies showed that the overexpression of TC21 were observed in breast cancers, esophageal and oral carcinomas, which closely associated with the early stages of tumorigenesis. In this study, we focus on the expression and function of TC21 in HCC. The overexpression of TC21 to some extent was found in 7 HCC cell lines and 1 immortalized human liver cell lines. Furthermore, by using fluorescence recovery after photobleaching technique (FRAP), we demonstrated that the TC21 localized both in nucleus and cytoplasm. Moreover, colony formation and ectopic tumor formation assay implicated that TC21 enhanced the toumorigenesis of HCC cells. In addition, after synchronization by thymidine (0.1 mM) or nocodazole (200 ng/ml) for 16 h, then reverted to the original status. Here we show TC21 promote SMMC-7721 HCC cells to earlier enter and more fast progressed G0/G1 checkpoint. Our study also demonstrated that TC21 enhanced tumorigenesis through activation of Akt and CDK kinases. But the mechanism of TC21 in Huh7 was significantly different from that in SMMC-7721 cells, which suggests there may exist different fuction and nechanisms of TC21 among various HCC cell lines.
