Abstract

The purpose of the experiment in human adenoid cystic carcinoma histology specimens to find and confirm the significance of vasculogenic mimicry structure to clarify its relationship with histological type and its expression. Observed by tumor stem cell markers associated protein expression of CD44v6 expression of CD133, of ABCG2 labeled specimens in the histological specimens of human adenoid cystic carcinoma, correlation analysis comparison angiogenesis mimicry between local recurrence to provide easy for this tumor a new interpretation. Morphology was observed again from the cellular level, specifically angiogenesis mimicry structure of the Department of SACC-83 in the presence of adenoid cystic carcinoma, and observe the cancer stem cell marker related expression of CD44v6, CD133, of ABCG2 in salivary adenoid cystic carcinoma cell line SACC-83 expression. VM generated mechanism lay the foundation for further tumor stem cell extraction and analysis. The method the experimental collection Second Hospital of Tianjin Medical surgical resection between January 1999 -2009 in March with the experimental conditions lacrimal gland adenoid cystic adenocarcinoma specimens of 33 cases. All specimens were re-sliced ??unified observed after HE staining confirmed the pathological diagnosis and classification. Comparative observation under an optical microscope after through CD31/PAS double stained slices, in the human lacrimal gland adenoid cystic adenocarcinoma specimens to locate and confirm the VM structure. The application of the two-step immunohistochemistry detected in 33 specimens of CD44v6, CD133 and ABCG2 expression. According to the the VM structure of the lacrimal gland adenoid cystadenocarcinoma observed, the application of statistical methods to compare their expression in different pathological type well related protein expression of CD44v6 VM expression group and the non-expression of the tumor stem cell marker CD133, of ABCG2 expression of any differences. By human salivary adenoid cystic carcinoma SACC-83 cell line from Beijing Medical University Stomatological Hospital, Li Shenglin Central Laboratory gifts) two-dimensional culture and three-dimensional culture, observation of vasculogenic mimicry and cancer stem cell marker protein expression of CD44v6, CD133, of ABCG2 SACC-83 in the expression in the cell lines. Results of to 1.33 specimens has been observed HE staining, and combined with the results of immunohistochemistry confirmed for the lacrimal gland adenoid cystic carcinoma, which entity type, 13 cases (39.39%), the adenoid - tubular type, 20 cases (60.61%). Observed through CD31/PAS double staining method found in 10 specimens VM structure, occurrence rate was 30.3%. Entity type 6 cases (46.2%), the adenoid - tubular type 4 cases (20.0%), the difference was not statistically significant (p = 0.346). VM 2. Observed by the adenoid cystic carcinoma cells surround formed tubular, fissure-like structure, within the structure of the lacunar visible erythrocytes, the structural integrity of these erythrocytes, and not around the presence of hemorrhage. Also observed part of the cytoplasm of tumor cells endocrine PAS positive substances and CD31-positive substance. VM expression group and the non-expression of CD44v6 expression was 1.57% and 2.51%, respectively, both have a statistically significant difference (p = 0.023). 3.33 specimens using immunohistochemical methods observed tumor stem cell-associated marker protein expression of CD44v6 expression of CD133, of ABCG2 were: CD44v6 expression rate was 54.5% (18/33). Entity-type positive rate of 76.9% (10/13), adenoid - tubular type positive rate of 40.0% (8/20), the difference was statistically significant (p = 0.037). CD133 positive expression rate was 57.6% (19/33), respectively, in the entity type and adenoids - tubular in 76.92% (10/13), 45% (9/20), the difference was not statistically significant (p = 0.070); expression product positioning in the cell membrane and cytoplasm, pale yellow to brown, in some cases at the same time expressed in the cytoplasm and nucleus. ABCG2 positive expression rate was 21.2% (7/33), respectively, in the entity type and adenoids - tubular in 30.8% (4/13), 15.0% (3/20), the difference was not statistically significant (p = 0.297); ABCG2 positive cells along blood vessels tendencies. In salivary adenoid cystic carcinoma cell line SACC-83 two-dimensional culture, visible cell annularly mesh growth was cavernous, three-dimensional culture, tubular growth. VM positive expression seen in three-dimensional culture. Tumor stem cell-associated protein in adenoid cystic carcinoma Department of SACC-83, visible expression of CD44v6 was 42.47 ± 7.37%, CD133 expression was 56.56 ± 10.27% of ABCG2 expression was 2.94 ± 1.23%. Conclusion 1. Lacrimal gland adenoid cystic carcinoma and human salivary gland adenoid cystic carcinoma cell line SACC-83 in both VM structure expression 2. Lacrimal gland adenoid cystic carcinoma pathological type VM expression has no direct impact CD44v6 and its sub-type-related; 3. lacrimal gland adenoid cystic carcinoma VM and CD44v6 expression related, prompted not only the VM with human lacrimal gland adenoid cystic carcinoma metastasis recurrence, and CD44v6 may affect the generation of the VM . 4 people lacrimal gland adenoid cystic carcinoma and salivary adenoid cystic carcinoma cell line SACC-83 different levels of expression of cancer stem cell marker proteins expression of CD44v6, CD133, of ABCG2, prompts the human lacrimal gland adenoid cystic carcinoma may the presence of tumor stem cells;
